扶正活血中药治疗放射性肺损伤 的实验研究

摘要:
目的 观察放射性肺损伤大鼠肺组织转化生长因子-β1(TGF-β1)表达水平及肺组织病理学特征,探讨扶正活血中 药对放射性肺损伤的防治作用及可能机制。方法 将56 只SD 雌性大鼠随机分为模型组、前期干预组、后期短程干预组及后期长程干预组,每组14 只,通过单次大剂量6MV-X线加速器胸部照射制成放射性肺损伤大鼠模型,分别不用及照射前2 周内、照射后1个月内、照射后3 个月内用扶正活血中药灌胃,于照射后3、6 个月分别随机处死各组大鼠7 只,取大鼠肺组织进行免疫组化检测TGF-β1蛋白表达强度及观察不同时相肺组织病理学特征。结果 各干预组大鼠肺组织TGF-β1蛋白表达强度明显低于模型组,放射性肺损伤病理学改变明显轻于模型组。结论 扶正活血中药具有抑制TGF-β1分泌,有效阻止或延缓肺纤维化的形成,对放射性肺损伤的防治具有一定作用。

Protective effects of Chinese medicine of Fuzheng Huoxue on radiation-induced lung injury in rats

Abstract:
Objective To investigate the effect of Chinese medicine of Fuzheng Huoxue on radiation-induced lung injury in rats. Methods Fifty six S-D rats were randomly allocated into irradiation group(n=14), early intervention group(n=14),short-term intervention group (n=14), long-term intervention group (n=14). The radiation lung injury was induced by irradiation on chest of rats with a single large dose using 6-MV linear accelerator. Chinese medicine of Fuzheng Huoxue was administrated by gavage at 2 weeks before irradiation(early intervention group), within 1 month or 3 months after irradiation(short-term or long-term intervention groups), respectively. The animals were sacrificed at the end of the 3 and 6 months after irradiation (7 in each batch). The histopathological changes of lungs was observed and the expression of TGF-β1 in lung tissue was detected with immunohistochemistry. Results The expression of TGF-β1 in intervention groups was significantly lower than that in model group. Compared to model group the histopathological changes of radiation-induced lung injury were significantly ameliorated in intervention groups. Conclusion Chinese medicine of Fuzheng Huoxue can alleviate or delay the radiation-induced lung injury in rats, which may be associated with the down-regulation of TGF-β1 expression in lung tissue.
