目的 通过研究过敏性休克小鼠模型致敏组和对照组中 IL-4、IL-6 和 IgE 的不同表达,进一步探讨过敏性休克的免 疫学发病机制,为人类过敏性休克的治疗提供新的思路。 方法 雄性昆明种小鼠 60 只随机分成致敏组 40 只和对照组 20 只。致敏组 小鼠于第 1 天腹腔注射致敏原卵蛋白(OVA)0.25ml,1 周后加强注射一次,饲养 3 周后尾静脉注射 OVA 0.5ml 诱发过敏性休克;对照 组以同等量的 PBS 缓冲液代替。以酶联免疫吸附(ELISA)法检测致敏组和对照组小鼠血清及肺泡灌洗液(BALF)中 IL-4、IL-6 和 IgE 的水平。 结果 成功建立过敏性休克小鼠模型。致敏组血清 IL-4、IL-6 和 IgE 的水平分别是对照组的 3.59、2.49、2.83 倍。致敏组 BALF 中 IL-4、IL-6 和 IgE 的水平分别是对照组的 2.75、1.82、1.58 倍。致敏组血清及 BALF 中 IL-4、IL-6 和 IgE 水平两两呈正相关 (均 P<0.01);对照组血清及 BALF 中 IL-4、IL-6 和 IgE 水平均无关(均 P >0.05)。 结论 血清及 BALF 中 IL-4、IL-6 水平升高提 示细胞因子可能参与过敏性休克发病过程。IL-4、IL-6 与 IgE 之间的相关性研究提示,IL-6 可能通过上调 IL-4 的表达在过敏性休克 的发病过程中发挥作用。 |

IL-6, IL-4 and IgE levels in serum and BALF of anaphylactic shock mice |

Objective To investigate IL-6, IL-4 and IgE levels in serum and bronchioalveolar lavage fluid (BALF) of mice with anaphylactic shock. Methods Sixty male KM mice were randomly divided into sensitized (n=40) and control groups (n=20).The mice in sensitized group were immunized by intraperitoneal injection of allergen at first day and then repeated after a week; allergic shock was induced by tail vein injection after three weeks later. The mice in control group were injected with equal volume of PBS buffer instead. The IL-6, IL-4 and IgE levels in serum and BALF were measured by ELISA. Results The mice model of anaphylactic shock was successfully established. Serum and BALF IL-6, IL-4 and IgE levels in sensitized group were increased by 3.59, 2.49, 2.83 times and by 2.75, 1.82, 1.58times, respectively. Serum and BALF levels of IL-6, IL-4 were positively correlated with those of IgE; and the IL-6 levels were also positively correlated with those of IL-4. In control group, there was no correlation of IL-6, IL-4 and IgE in serum and BALF with each other. Conclusion Increased serum and BALF levels of IL-6, IL-4 in sensitized mice indicate that the two cytokines are likely to participate in the pathogenesis of anaphylactic shock in mice. |
