重组IL-4R蛋白对SD大鼠过敏性哮喘干预作用的研究

摘要:
目的探索重组IL-4R蛋白对过敏性哮喘动物模型的干预作用。方法选择6周龄SD大鼠60只,采用安全随机分组法分成哮喘模型组、IL-4R组、布地奈德组和对照组,每组15只。哮喘模型组在腹部选取3处于皮下注射10%鸡卵清白蛋白(OVA)混合液1ml(OVA100mg、氢氧化铝100mg及灭活的百日咳杆菌5×109个)进行致敏,1周后用上述同样药物再进行致敏,第2次致敏1周后将大鼠分别放入不完全封闭的箱中,用5%OVA与0.9%氯化钠混合溶液,连续10d超声雾化激发30min。IL-4R组和布地奈德组同法造模,但每次雾化激发前30min,IL-4R组腹腔注射重组IL-4R蛋白稀释液(400滋g/d)。布地奈德组腹腔注射布地奈德稀释液(400滋g/d),对照组用0.9%氯化钠溶液连续10d超声雾化激发30min。取4组大鼠肺组织作HE染色的光镜病理切片,观察肺组织病理变化。建立间接ELISA方法,检测实验中4组大鼠肺泡灌洗液中的IL-4和IFN-γ的含量。结果哮喘模型组大鼠肺组织有大量嗜酸性粒细胞浸润,对照组大鼠肺组织几乎无嗜酸性粒细胞浸润,IL-4R组和布地奈德组大鼠肺组织嗜酸性粒细胞浸润明显少于哮喘模型组。IL-4R组和布地奈德组肺泡灌洗液中IL-4和IFN-γ的含量明显高于哮喘模型组,差异均有统计学意义(均P<0.05)。结论研究结果提示IL-4R蛋白对过敏性哮喘有良好的干预作用。

Intervention of allergic asthma with recombinant IL-4R in rats

Abstract:
Objective To investigate the effect of recombinant IL-4R on allergic asthma in rats. Methods Sixty SD rats in 6 wk of age were randomly divided into model group, IL-4R group, budesonide group (as positive control) and control group with 15 in each. Asthma model was induced with subcutaneous injection of ovalbumin (OVA). Rats in IL-4R group,budesonide group and control group were given intraperitoneal injection of recombinant IL-4R (400滋g/d), budesonide (400滋g/d) or normal saline, respectively. The pathological changes of lung tissues were observed with HE staining, the contents of IL-4 and IFN-γ in alveolar lavage fluid were detected by indirect ELISA method. Results Large amount of eosinophil infiltration in the lung tissue was observed in asthma model group, which were significantly attenuated in IL-4R budesonide groups. The contents of IL-4 and IFN-γ in alveolar lavagefluid of IL-4R and budesonide groups were significantly lower than those of asthma model group (P< 0.05). Conclusion The results suggest that recombinant IL-4R can effectively intervene allergic asthma in rats.
