目的探讨FTY720对颅脑损伤大鼠海马组织神经细胞凋亡的抑制作用。方法30只大鼠随机分成假手术组、模型组和治疗组,每组10只,采用改进的Feeney自由落体损伤装置建立大鼠颅脑损伤模型,假手术组大鼠切开头皮,颅骨钻孔后缝合头皮,不作外力打击。治疗组大鼠按1mg/kg剂量予FTY720腹腔注射,其他组大鼠腹腔注射0.9%氯化钠溶液1ml。24h后断头处死大鼠,分离海马组织,采用TUNEL法检测凋亡神经细胞,Western-blotting检测pro-caspase3/9蛋白表达,四肽荧光底物法检测caspase-3/9蛋白活性。结果模型组大鼠海马组织凋亡神经细胞比例、pro-caspase3/9蛋白表达和caspase-3/9蛋白活性较假手术组显著升高,而治疗组大鼠海马组织凋亡神经细胞比例、pro-caspase3/9蛋白表达和caspase-3/9蛋白活性较模型组显著下降。结论FTY720可显著抑制颅脑损伤大鼠海马组织神经细胞凋亡,具有脑保护作用。 |

Inhibitory effect of FTY720 on cell apoptosis of neurons in rats with experimental traumatic brain injury |

Objective To investigate the inhibitory effect of FTY720 on cell apoptosis in hippocampus of rats with experimental traumatic brain injury. Methods Thirty rats were randomly divided into sham-operation group, model group and treatment group with 10 in each. Modified Feeney free fall injury device was used to induce craniocerebral injury in rats of model and treatment groups. Rats in sham-operation group underwent scalp incision, cranial drilling and scalp suture, but no traumatic impact. Rats in treatment group were administrated intraperitoneally with 1 mg/kg FTY720. Rats in other two groups were given an intraperitoneal injection of 1 mL normal saline. After 24 hours, rats were sacrificed by decapitation and their hippocampus tissues were dissected. TUNEL staining, Western-blotting and four peptide fluorescence substrate method were used to determine the apoptotic neuronal cells, expressions of pro-caspase 3/9 proteins and activities of caspase 3/9 proteins, respectively. Results Compared with sham-operation group, the percentage of apoptotic neuronal cells, expressions of pro-caspase 3/9 proteins and activities of caspase 3/9 proteins were significantly elevated in the hippocampus of model group; while these indicators in treatment group were significantly lower than those of model group. Conclusion FTY720 can markedly suppress cell apoptosis of hippocampus neurons in rats after trauma brain injury and shows cerebral protective effect. |
