红豆杉多糖对免疫功能低下小鼠细胞免疫功能的影响

摘要:
目的探讨红豆杉多糖增强免疫低下小鼠免疫功能的机制。方法将48只昆明小鼠按随机数字表法分为正常对照组,氢化可的松模型组,红豆杉多糖低、中、高剂量组及猴头菇多糖阳性对照组,每组8只。除正常对照组外,其余各组小鼠皮下注射氢化可的松0.2mg/kg,隔天1次,共14d,制备小鼠免疫功能低下模型。正常对照组和氢化可的松模型组予0.9%氯化钠溶液10ml/kg灌胃,红豆杉多糖低、中、高剂量组分别予等量不同浓度10、20、40mg/ml红豆杉多糖溶液灌胃,猴头菇多糖阳性对照组予等量浓度为1.1mg/ml猴头菇多糖溶液灌胃,均1次/d,连续14d。流式细胞术检测小鼠血CD4+、CD8+细胞百分比,ELIAS法检测血清IL-4水平,测量小鼠胸腺指数。结果与正常对照组比较,氢化可的松模型组CD4+、CD8+细胞百分比均明显降低(P<0.01),红豆杉多糖低、中、高剂量组与氢化可的松模型组对比,CD4+、CD8+细胞百分比均有所增加(均P<0.05),其中红豆杉多糖中剂量组能降低CD4+/CD8+比值(P<0.05)。与正常对照组比较,氢化可的松模型组血清IL-4水平明显下降,差异有统计学意义(P<0.01);红豆杉多糖低、中、高剂量组IL-4水平均高于氢化可的松模型组(P<0.05或0.01)。氢化可的松模型组与正常对照组比较,胸腺指数明显降低(P<0.01)。红豆杉多糖低、中、高剂量组及猴头菇多糖阳性对照组与氢化可的松模型组比较,胸腺指数均有所增加(P<0.05),其中红豆杉多糖中剂量组及猴头菇多糖阳性对照组均明显增高(P<0.01)。结论红豆杉多糖增强细胞免疫的机制它可能主要通过调节细胞免疫来增强免疫低下小鼠的免疫功能。

Effects of taxus polysaccharide on cellular immunity of immunosuppressed mice

Abstract:
Objective To investigate the effect of taxus polysaccharide on cellular immunity of immunosuppressed mice. Methods Forty-eight male Kunming mice were divided into 6 groups. Five groups of mice were administered with hydrocortisone (Hy) (0.2mg•kg-1 q.o.d x14d s.c) to induce immunosuppression, 3 of these groups received taxus polysaccharide treatment (10mg/ml, 20mg/ml, 40mg/ml ), 1 group received polysaccharide of hericium erinaceusthe (HEP), 1 group received normal saline (NS). Serum CD4+, CD8+ levels were detected by flow cytometry, and the thymus index was measured. Results The number of CD4+T cells, CD8+T cells were significantly reduced in Hy-groups compared with normal group, but in high-dose, and middle-dose the number of CD4+T cells, CD8+T cells was significantly increased (P<0.01), the CD4+ / CD8+ ratio was decreased in middle dose group compared with model group (P <0.05). Compared with control group, thymus index was elevated in treatment groups(P<0.05), which was more markedly in the middle dose group(P<0.01). Conclusion Taxus polysaccharide can improve the cellular immunity in immunodepressed mice.
