miR-29a在系统性红斑狼疮患者血浆中的表达及意义

摘要:
目的 分析microRNA-29a(miR-29a)在系统性红斑狼疮(SLE)患者血浆中的表达水平及与SLE临床特征的相关性。方法建立检测miR-29a的实时荧光定量PCR(RT-qPCR)方法,分析48例SLE患者和20例健康体检者血浆中miR-29a的表达水平,ROC曲线评估其作为SLE诊断标记物的意义,统计分析其表达与SLE患者临床特征的相关性。结果与健康体检者比较,SLE患者血浆miR-29a表达水平明显上调(Z=3.476,P=0.0009);血清miR-29a诊断SLE的ROCAUC为0.794(95%CI:0.689~0.898),具较好诊断SLE的效能;抗β2GP1抗体>20RU/ml患者的血浆miR-29a水平明显升高(P=0.033),Pearson相关分析显示,SLE患者血浆miR-29a表达水平与抗β2GP1抗体水平呈正相关(r=0.584,P=0.001),而与不同SLEDAI评分及其他SLE活动相关指标等差异均无统计学意义(均P>0.05)。结论miR-29a在SLE患者血浆中的表达水平明显上调,且血浆miR-29a上调与SLE患者自身抗体水平相关,其调节机制尚待进一步的研究。

Expression of miR-29a in plasma of patients with systemic lupus erythematosus and its clinical significance

Abstract:
Objective To investigate the expression of microRNA-29a (miR-29a) in plasma of patients with systemic lupus erythematosus(SLE) and its clinical significance. Methods The expression of plasma miR-29a was detected with real-time quantitative polymerase chain reaction(qRT-PCR)in 48 SLE patients and 20 healthy subjects. The value of miR-29a for diagnosis of SLE was assessed by the receiver operating characteristic (ROC) curve. The relationship between the plasma miR-29a levels and clinical characteristics was analyzed. Results Compared with the healthy controls, the expression of miR-29a in plasma of patients with SLE was significantly higher (Z=3.476, P=0.0009). The area under the curve (AUC) of ROC was 0.794 (95% CI: 0.689-0.898) . Patients with positive anti-β2GP1 antibody showed higher miR-29a levels in plasma than those with negative anti-β2GP1 antibody (P =0.033). In addition, Pearson analysis indicated that the plasma level of miR-29a was positively correlated with the level of anti-β2GP1 antibodies (r=0.584, P=0.001). However, the SLEDAI score and other clinical indicators of SLE activation were not significantly correlated with the plasma level of miR-29a. Conclusion The expression of miR-29a in the plasma of SLE patients is up-regulated, which were associated with the levels of several auto-antibodies.
