TGF-βR Ⅱ基因在乳腺癌中的表达及临床意义

摘要:
目的 检测 TGF-βRⅡ基因在乳腺癌及癌旁正常乳腺组织中的表达水平,并探讨其在乳腺癌中的临床意义。 方 法 选取初治乳腺癌患者病理标本 65 例,分别运用 RT-PCR 和 Western blot 检测 TGF-βRⅡ的 mRNA 及蛋白在乳腺癌组织和癌 旁正常乳腺组织中的表达,统计分析乳腺癌组织中 TGF-βRⅡ的表达与患者临床病理特征的关系。 结果 乳腺癌组织中 TGF-βR Ⅱ 的 mRNA 表达水平低于癌旁正常乳腺组织(0.4352±0.1623 vs 0.5218±0.1236,P<0.05),同时其编码的 TGF-βRⅡ蛋白表达水 平也低于癌旁正常乳腺组织(0.2165 vs 0.3725,P<0.05)。TGF-βRⅡ蛋白表达情况与原发肿瘤大小、临床分期有关,肿瘤越小、临床 分期越早的患者中 TGF-βRⅡ蛋白高表达的比例越高(均 P<0.05),而与患者年龄、淋巴结转移、组织学类型及分级等无关(均 P > 0.05)。 结论 TGF-βRⅡ基因在乳腺癌组织中呈低表达,且与肿瘤大小、临床分期有关,检测 TGF-βRⅡ的表达对评估乳腺癌生物 学行为及预后具有较重要的临床意义。

Expression of TGF-βR Ⅱ in breast cancer and its relation to clinicopathological features

Abstract:
Objective To study the expression of transforming growth factor beta receptor II (TGF-βRⅡ) in breast cancer, and to investigate its relationship with and clinicopathological features of the cancer. Methods The expression of TGF-βRⅡ mRNA and protein was detected by RT-PCR and Western blot in 65 cases with breast cancer and their adjacent tissues, and the relationship between the expression of TGF-βRⅡ and clinicopathological features was analyzed. Results The expression of TGF-βR Ⅱ mRNA in cancer tissues was lower than that in adjacent normal breast tissue (0.4352 ±0.1623 vs 0.5218±0.1236, P<0.05), and the expression levels of TGF-βRⅡ protein showed very similar trends (0.2165 vs 0.3725, P< 0.05). The expression of TGF-βRⅡ in breast cancer was negatively correlated with tumor size and clinical stage (P<0.05), but not correlated with age, lymph node metastasis, histological types and clinical grades (P >0.05). Conclusion In breast cancer the expression of TGF-βRⅡ is down-regulated, which is correlated with tumor size and clinical stage of the cancer.
