支气管哮喘患者白三烯受体1基因MicroRNA的表达

摘要:
目的探讨支气管哮喘患者外周血细胞白三烯受体-1(CysLTR1)基因MicroRNA的表达及临床意义。方法选择支气管哮喘急性发作患者17例(哮喘发作组)、支气管哮喘诊断明确且临床症状完全缓解3个月以上患者15例(哮喘缓解组)和健康体检者20例(正常对照组),采用RT-PCR技术检测外周血细胞CysLTR1基因MicroRNA的表达,并比较分析3组检测结果。结果CysLTR1基因MicroRNA在正常对照人群和支气管哮喘患者中均有表达,但表达水平不同:哮喘发作组、哮喘缓解组和正常对照组CysLTR1/GAPDH分别为2.4059±0.1983、2.1800±0.2210和1.2300±0.1174,与正常对照组相比,哮喘发作组和哮喘缓解组CysLTR1基因MicroRNA的表达均明显增高(均P<0.05);哮喘发作组与哮喘缓解组比较,差异无统计学意义(P>0.05)。结论哮喘患者CysLTR1基因MicroRNA高表达可能与哮喘发生或急性发作相关。

Abstract:
Objective To investigate the expression of cysteinyl leukotrienes receptor 1 (CysLTR1) MicroRNA in peripheral blood mononuclear cell(PBMC) of patients with asthma. Methods Seventeen patient with asthma of acute attack, 15 patients with remission stage of asthma and 20 healthy individuals were enrolled in the study. CysLTR1 MicroRNA in PBMC was detected by RT-PCR. Results The expression of CysLTR1 MicroRNA was significantly higher in patients with asthma of acute attacks and with remission than in controls (CysLTR1 /GAPDH 2.4059 ±0.1983 and 2.1800±0.2210 vs 1.2300 ±0.1174, P<0.05). But there was no significant difference between patients with acute attacks and those with remission (P >0.05). Conclusion The up-regulated CysLTR1 MicroRNA in PBMC of asthma patients may be associated with occurrence or acute attack of the disease.
