		     | 目的分析miRNA-133b表达水平的改变与胃癌临床病理特征之间的关系,探讨miRNA-133b在胃组织中表达的意义。方法选择手术治疗的胃癌患者肿瘤组织63例为研究对象,取切缘正常胃黏膜为对照,设计针对miRNA-133b的特异性引物,应用定量PCR方法检测胃癌组织中miRNA-133b表达水平的变化。结合临床病理资料对miRNA-133b的表达水平与肿瘤病理类型及临床病理分期之间的关系进行分析。检测胃癌细胞株中miRNA-133b的表达水平变化。结果miRNA-133b在胃癌组织中表达水平明显低于正常胃黏膜组织[ΔCt:(-6.94±3.319)vs(-4.98±2.846),P<0.001]。其中45例miRNA-133b下调≥2.0倍,占71%;6例上调≥2.0倍,占10%;12例无明显变化,占19%。miRNA-133b与肿瘤病理类型、肿瘤TNM分期无明显相关性(P>0.05)。与正常胃黏膜细胞相比,在胃癌细胞株中miRNA-133b表达明显下调(P<0.05)。结论在胃癌组织及胃癌细胞中,miRNA-133b的表达水平均有不同程度的降低,但其下降程度和比例与临床特征无明显相关性。miRNA-133b的表达水平下调可能与促进肿瘤细胞增殖有关。 |

                | Objective To investigate the expression of miR-133b in  gastric  cancer  and  its  clinicopathological significance. Methods Sixty three specimens of primary gastric cancer and corresponding pericancerous gastric tissue were collected. The level of miRNA-133b was examined using quantitative real-time PCR assay (qPCR); the correlation between miRNA-133b level and clinicopathologic features were analyzed. The expression of miR-133b in 5 human gastric cancer cell lines was also examined by qPCR. Results The mean expression level of miRNA-133b was down-regulated in gastric cancer tissues compared with the adjacent tissues[ΔCt: (-6.94 ± 3.319) vs (-4.98 ± 2.846), P<0.001]. Among 63 pairs gastric cancer tissues, 45 cases showed down-regulation of miRNA-133b >2 folds, 12 cases had no difference, 6 cases displayed up-regulation >2 folds. No significant associations were detected between the level of miRNA-133b expression and TNM stage in gastric cancer (P >0.05). miRNA-133b was also markedly down-regulated in gastric cancer cell lines. Conclusion miRNA-133b expression  is down-regulated in  gastric  cancer tissue and human gastric cell lines.  There  is  no association   of
