目的 建立兔耳创面模型,研究纳米氧化锌(Nano-ZnO)对创面瘢痕增生形成的抑制作用。 方法 8 只新西兰大白兔 随机分成两组,分别在两组兔耳腹侧制作直径约 1.0cm 的创面,实验组在创面喷洒 Nano-ZnO,对照组创面喷洒 0.9%氯化钠溶液,每隔 1d 喷洒 1 次。术后 15d 当创面全部愈合后,继续在两组兔耳创面的愈合瘢痕组织上喷洒 Nano-ZnO 及 0.9%氯化钠溶液。术后 22、39 及 54d 搜集创面的瘢痕组织,利用游标卡尺测量瘢痕厚度,同时行组织形态学观察,并以免疫组化半定量检测Ⅰ型胶原及Ⅲ型胶原的组织沉 积情况。 结果 大体形态学观察,实验组的瘢痕增生明显轻于对照组;组织病理学染色显示,实验组的瘢痕厚度明显薄于对照组,胶原纤 维束走向更接近于正常皮肤。免疫组化染色显示,随时间推移实验组Ⅰ型胶原表达逐渐减弱,对照组表达逐渐增强;实验组Ⅲ型胶原表达 则逐渐增强,对照组则呈减弱趋势;后期实验组Ⅲ型胶原表达高于对照组。术后 22、54d 时各组之间Ⅰ型胶原、Ⅲ型胶原的表达比较均有 统计学差异(均 P<0.05)。 结论 Nano-ZnO 可以促进创面愈合,具有降低兔耳腹侧增生性瘢痕中Ⅰ型胶原表达,促进Ⅲ型胶原表达的 作用,从而能够减轻瘢痕增生,促使已形成的增生性瘢痕软化、吸收。 |

Inhibitive effect of Nano-ZnO on hypertrophic scar in rabbits |

Objective To investigate the effect of Nano-ZnO on hypertrophic scar in rabbits after injury. Methods Eight New Zealand white rabbits were randomly assigned into Nano-ZnO group and control group with 4 rabbits in each group. Full-thickness wounds with 1.0cm in diameter were made over the ventral surface of rabbit ears. Nano-ZnO was applied on the wounds of rabbit ears twice a day in Nano-ZnO group; while normal saline was applied in control group. The expression of type Ⅰ and Ⅲ collagen in hypertrophic scar of rabbits ears were determined by immunohistochemical staining at different time points. Results Excessive dermal scars on rabbit ears appeared in both groups, but the scars were attenuated in Nano-ZnO group. The expression of typeⅠcollagen in hypertrophic scar of rabbit ears after injury in Nano-ZnO group was lower than that in control group; while the expression of type Ⅲ collagen was higher in Nano-ZnO group than that in control group. Conclusion The Nano-ZnO can reduce the hypertrophic scar formation, accelerate softening and vanishing process of excessive scar in rabbit ears. |
