宫颈癌组织中miR-21和Stat3的表达及相关性研究

摘要:
目的探讨微小RNA-21(miR-21)和信号转导及转录活化因子-3(Stat3)在宫颈癌中的表达情况及临床意义。方法采用实时荧光定量PCR检测48例宫颈癌及20例正常宫颈组织中miR-21和Stat3mRNA的表达,免疫组化Envision法检测宫颈癌及正常宫颈组织中Stat3蛋白表达,分析miR-21和Stat3表达的相关性及其与宫颈癌患者临床病理特征的关系。结果miR-21和Stat3mRNA在宫颈癌组织中的表达均显著高于正常宫颈组织(均P<0.05),且miR-21和Stat3mRNA的高表达呈正相关(r=0.631,P<0.05)。Stat3蛋白在癌组织中的表达阳性率为79.17%(38/48)。Stat3的高表达和肿瘤低分化及有淋巴结转移有关(均P<0.05)。结论miR-21在宫颈癌中的表达升高,可能和宫颈癌的增殖、侵袭转移有关。

Expression of miR-21 and Stat3 in cervical cancer and their clinicopathological significance

Abstract:
Objective To investigate the expression of microRNA-21 (miR-21) and Stat3 in cervical cancer and their clinicopathologic significance. Methods Stem-loop real-time RT-PCR was used to examine the expression of miR-21 and Stat3 in 48 specimens of cervical cancer and 20 specimens of normal cervical tissue. Immunohistochemistry was used to examine Stat3 protein expression in the tumor tissue. The association of miR-21 and Stat3 expression with clinicopathologic features was analyzed. Results The expression of miR-21 and Stat3 in cervical cancer tissues were significantly higher than that of normal tissues (P <0.05). The expression rate of Stat3 in cervical cancer tissue was 79.17% (38/48). The expression of miR-21 was positively correlated with Stat3 (r=0.631, P<0.05). The expression of Stat3 was correlated with lower differentiation, lymph node metastasis in cervical cancer (P<0.05). Conclusion The expressions of miR-21 and Stat3 are up-regulated in cervical cancer, which may be involved in the development and progression of cervical cancer.
