目的 分析胃癌组织特征长链非编码RNA(lncRNA)表达谱,并探讨长链非编码RNA UCA1 在胃癌中的表达及效
应。方法 采用基因芯片检测6 例胃癌患者的肿瘤组织及癌旁正常组织LncRNA表达水平,分析胃癌组织特征lncRNA 表达谱,进一步通过定量PCR 检测40 例胃癌及其对应的癌旁正常组织中UCA1的表达并分析淋巴结转移、肿块大小、细胞分化程度及病理分期等临床病理特征的关系。在此基础上,利用siRNA 下调AGS 胃癌细胞内UCA1 表达水平,利用CCK-8 试剂盒检测UCA1干预对胃癌细胞增殖能力的影响。结果 LncRNA 基因芯片检测到胃癌组织1 021条差异>2倍的lncRNA(P<0.05),定量PCR 验证明显差异表达的UCA1在胃癌组织中的表达显著高于正常组织(P<0.01)。此外,临床病理相关分析显示,UCA1表达水平还与胃癌的细胞分化程度及病理分期相关。分化低的胃癌组织中UCA1表达水平明显高于中、高分化的胃癌组织(P<0.05);病理分期Ⅲ/Ⅳ期胃癌组织的UCA1 表达水平明显高于Ⅰ/Ⅱ期的标本(P<0.05)。此外,下调UCA1 胃癌细胞表达可明显抑制细胞增殖。结论 异常表达的lncRNA 参与了胃癌的发生、发展,其中肿瘤组织上调表达的UCA1是胃癌重要的促癌基因。 |

Objective To investigate the profiles of long non-coding RNA (lncRNA) expression in gastric cancer tissue.Methods The expression of lncRNAs was detected with microarray assay in 6 samples of gastric tumor and matched adjacent tissues. The profiles of lncRNAs in gastric cancer tissues were identified. The UCA1 expression was evaluated by real-time reverse transcription polymerase chain reaction (RT-PCR) in 40 gastric tumor samples and matched adjacent tissues.The correlations of UCA1 expression with lymph node metastasis, tumor size, cell differentiation, pathological stage were further analyzed. Tumor cell proliferation was assessed following siRNA knockdown of UCA1 by using the CCK 8 kits. Results A total of 1 021 lncRNA expressed in gastric tumor samples >2 folds than matched adjacent tissues were identified (P<0.05) using microarray assay. RT-PCR showed that the expression of UCA1 was significantly highly in gastric cancer than that in adjacent tissues
(P<0.0001). High UCA1 expression was associated with cell differentiation and pathological stage of gastric cancer. UCA1 expression in low-differentiated gastric cancer was significantly higher than that in with high- differentiated cancer (P=0.031). In addition, UCA1 level in stage Ⅲ/Ⅳ gastric cancer was significantly higher than that with stage I/II cancer (P=0.013). Furthermore,
inhibition of UCA expression suppressed the proliferation of gastric cancer cells. Conclusion Abnormal expression of LncRNA may be involved in the development of gastric cancer. Up-expression of UCA1 in tumor tissue might act as an oncogene and promote the development of gastric cancer. |
