目的探讨MAL基因对大肠癌筛查及早期诊断的价值。方法选取10例正常大肠组织作为对照,应用RT-PCR、甲基化特异性PCR、Westernblot方法分别检测并比较大肠癌组织(40例)及相应癌旁组织(40例)中MALmRNA、蛋白的表达情况以及MAL基因启动子甲基化状态。结果大肠癌组织中MALmRNA、蛋白的表达阳性率(10.0%、20.0%)均低于其在癌旁组织(75.0%、85.0%)和正常大肠组织(100.0%、100.0%)中的表达阳性率(均P<0.05),而大肠癌组织中MAL基因启动子甲基化率(90.0%)均高于其在癌旁组织(40.0%)和正常大肠组织(0.0%)中的甲基化率(均P<0.05)。结论大肠癌组织中MAL基因或可作为大肠癌筛查及早期诊断的高潜力分子标志物。 |

Objective To investigate the expression of MAL gene and the status of its promoter methylation in colorectal cancer. Methods The expression of MAL mRNA and protein and status of promoter methylation in 40 samples of colorectal carcinoma and corresponding adjacent tissues were detected by reverse transcription-polymerase chain reaction (RT-PCR), Western blot and methylation specific PCR, respectively. Ten samples of normal colorectal tissue were used as controls. Results
The expression rates of MAL mRNA and protein in colorectal cancer tissues (10.0%, 20.0%) were lower than those in adjacent tissues (75.0%, 85.0%) and normal colorectal tissues (100.0%, 100.0%) (all P<0.05). The methylation rate of MAL promoter in colorectal cancer tissues (90.0%) was higher than that in adjacent tissues (40.0%) and normal colorectal tissues (0.0%) (all P< 0.05). Conclusion MAL gene is down-regulated in colorectal carcinoma, suggesting that it might be used as a molecular marker
for screening and early diagnosis of colorectal cancer. |
