SCID小鼠永久性左侧大脑中动脉阻塞模型的制备及评价

摘要:
目的 研究严重免疫缺陷(SCID)小鼠永久性左侧大脑中动脉闭塞(L-MCAO)模型的制备及评价。 方法 将 SCID 小鼠随机分为假手术组和 L-MCAO 组各 15 只。于 SCID 小鼠嗅束外缘电凝并切断左侧大脑中动脉(L-MCA),制作 L-MCAO 模型。 使用激光散斑血流仪实时监测小鼠脑血流量,并计算脑血流量比值;术后在小鼠麻醉清醒后 24h 进行神经功能学评分;术后 2 周后取 小鼠大脑行 2,3,5- 氯化三苯基四氮唑 (TTC) 染色和二氨基联苯胺 (DAB) 染色,并计算脑梗死面积百分比。 结果 术后 60min L-MCAO 组小鼠脑血流量比值基本稳定在 0.47±0.55,神经功能学评分为(1.95±0.76)分,脑梗死面积百分比为(18.47±1.92)%;假 手术组及建模失败的小鼠未见梗死区域。 结论 SCID 小鼠永久性 L-MCAO 模型在一定程度上可以模拟人类脑梗死,制作成功率高, 在临床脑梗死观察和药物研究方面具有较高的应用价值。

Establishment of permanent left middle cerebral artery occlusion model in SCID mice

Abstract:
Objective To establish a permanent left middle cerebral artery occlusion (L-MCAO) model in severe combined immunodeficiency (SCID) mice. Methods Thirty SCID mice were randomly divided into sham operation group and L-MCAO group with 15 in each group. The left middle cerebral artery occlusion was induced by electrocoagulation in SCID mice of L-MCAO group. The cerebral blood flow was monitored by laser Doppler, and the ratio of cerebral blood flow was calculated. The neurological deficit score was assessed 24h after operation. The animals were sacrificed two weeks after the operation; the brain tissues were stained with 2, 3, 5-triphenyltetrazolium chloride ( TTC) and diaminobenzidine (DAB), and the percentage of cerebral infarction area was calculated. Results The ratio of cerebral blood flow in L-MCAO group was 0.47 依0.55, the neurological deficit score was 1.95±0.76, and the percentage of cerebral infarction area was (18.47±1.92)%. While there was no cerebral infarction lesion in the sham group and model failed mice. Conclusion The permanent L-MCAO model of SCID mice has been established with high success rate, which simulates human cerebral infarction to a certain extent, and may be used for research on cerebral infarction and drug development.
