槐耳清膏联合顺铂改变胃癌SGC7901细胞周期分布的实验研究

摘要:
目的观察槐耳清膏联合顺铂(DDP)作用于人胃腺癌SGC7901细胞后细胞周期的变化,并探讨其可能的作用机制。方法通过细胞流式实验,观察不同浓度槐耳清膏、DDP及两药联合作用于胃癌SGC7901细胞24h后细胞周期的变化情况,并通过Westernblot检测不同浓度的槐耳清膏、DDP及两药联合作用于胃癌SGC7901细胞后,细胞周期相关蛋白ATM、H2AX、cdc-2、CyclinB1等的表达情况。结果细胞流式实验结果显示,槐耳清膏能够阻滞SGC7901细胞于G2/M期,DDP能够阻滞SGC7901细胞于S期,而两药联用后S期和G2/M期都有阻滞。Westernblot实验结果显示,槐耳清膏、DDP均可上调p-ATM、γ-H2AX蛋白表达,同时下调CyclinB1蛋白表达,且两药联用后效果更加明显。结论槐耳清膏、DDP能够通过调节细胞周期相关蛋白的表达改变细胞周期,抑制细胞增殖,与槐耳清膏联用可提高DDP对胃癌的疗效。

Abstract:
Objective To investigate the combined effects of fungi Trametes robiniophila murr (Huaier) extracts and cis-platin (DDP) on cell cycle of gastric cancer cell line SGC7901in vitro and the underline mechanism. Methods Human gastric cancer SGC7901 cells were treated with Huaier and/or DDP in vitro. The cell cycle was examined by flow cytometry; and the expression of cell cycle-related proteins was detected by Western blot. Results The ratio of cells in G2/M phase of SGC7901 cells treated with Huaier were increased in a dose-dependent manner compared with control group; and the ratio of cells in S phase in DDP-treated groups were increased also in a dose-dependent manner. The cells arrested at G2/M and S phases were markedly increased in Huaier and DDP combined treatment groups. The expression of CyclinB1 was down-regulated and expression of p-ATM,酌-H2AX was up-regulated in Huaier and/or DDP treatment groups. Conclusion Huaier and DDP can change cell cycle through up-regulation of p-ATM, 酌- H2AX expression and down-regulation of CyclineB1 expression in human gastric cancer SGC7901 cells.
