目的探究中药经验方心肌1号对心肌损伤大鼠基质细胞衍生因子-1(stromalcellderivedfactor-1,SDF-1)的调节作用及骨髓干细胞向心肌组织归巢分化的影响。方法制作心肌损伤大鼠模型30只,按随机数字表法分为中药(心肌1号)组、中药(心肌1号)+AMD3100(SDF-1抑制剂)组和生理盐水组。1次/d中药或生理盐水灌胃,AMD3100腹腔注射,共4周。测定治疗前后3组大鼠的体重、左心室射血分数(LVEF)、外周血及心肌匀浆中SDF-1水平(ELISA法)及心肌组织中SDF-1mRNA表达(RT-PCR法),HE染色观察心肌组织病变,免疫荧光染色观察心肌组织中的新生细胞。结果治疗4周后,中药组大鼠的体重和LVEF高于中药+AMD3100组和生理盐水组。中药组与中药+AMD3100组大鼠的外周血及心肌匀浆中SDF-1水平、心肌组织SDF-1mRNA水平无统计学差异,均高于生理盐水组(P<0.01)。中药组心肌损伤部位炎症细胞稀少,组织修复明显,并可见较多呈均匀分布的心肌样新生细胞,而中药+AMD3100组和生理盐水组炎症细胞浸润明显,罕见新生细胞。结论心肌1号能提高心肌损伤大鼠的心功能,其机制可能与其上调SDF-1的表达,进而促进骨髓干细胞向心肌组织的归巢分化有相关。 |

Effect of TCM docoction Myocardial-1on SDF-1 expression and cadiac homing in rats with myocardial injury |

Objective To investigate the effect of traditional Chinese medicine (TCM) decoction Myocardial-1 on the expression of stromal cell derived factor-1 (SDF-1) and the cardiac homing and differentiation of bone marrow stem cells in rats with myocardial injury. Methods Myocardial injury was induced in 30 rats, and the rats were randomized into TCM group, TCM+AMD3100 group and NS group with 10 in each group. The Myocardial-1 decoction (TCM and TCM+AMD group) or normal
(NS group) were administrated by gavage, q.d,AMD3100 was administrated by intraperitoneal injection, q.d for 4 weeks
(TCM+AMD group). The body weight was recorded, left ventricular ejection fraction (LVEF) was examined, SDF-1 expression in serum and cardiac tissue was measured by ELISA, SDF-1 mRNA in cardiac tissue was detected by RT-PCR, before and 4 weeks after treatment. HE staining and immunofluorescence-triple-label staining were adopted to observe the myocardial pathological changes and the regenerated cells. Results After 4 weeks of treatment the expression levels of SDF-1 in serum and tissue, SDF-1 mRNA in tissue of TCM group and TCM+AMD3100 group were significantly higher than those in NS group (P< 0.01); while there was no significant difference between TCM group and TCM+AMD3100 group. The body weight and LVEF in TCM group were higher than those in TCM+AMD3100 group. The necrotic lesions were attenuated in TCM group with fewer inflammatory cells and more cardiomyocyte-like cells. While the TCM+AMD3100 group and NS group had more severe inflammation and fewer regenerated cells. Conclusion TCM decoction Myocardial-1 can improve the cardiac function of rats with myocardial injury, which may be related to up-regulation of SDF-1 expression resulting in the cardiac homing and differentiation of bone marrow stem cells. |
