| 目的探讨血管生成素-2(Ang-2)及其受体内皮细胞特异性酪氨酸激酶受体(Tie-2)在宫颈癌组织中发生、发展和临床病理学特征的关系。方法采用免疫组织化学、RT-PCR及Westernblot分别检测Ang-2及Tie-2在60例正常宫颈及宫颈癌组织中的组织定位、基因差异表达及蛋白差异表达;采用Spearman法分析Ang-2及Tie-2的表达相关性。结果宫颈癌组织中Ang-2及Tie-2的阳性表达率明显高于正常宫颈组织,差异均有统计学意义(均P<0.05)。Ang-2及Tie-2在淋巴结发生转移或临床分期Ⅲ~Ⅳ级的患者中的表达明显高于未发生淋巴结转移或临床分期Ⅰ~Ⅱ级的患者(均P<0.05)。宫颈癌组织中Ang-2及Tie-2mRNA及蛋白表达水平均明显高于正常宫颈组织,差异均有统计学意义(均P<0.01)。Tie-2与Ang-2在宫颈癌组织中的表达呈正相关(r=0.8479,P<0.01)。结论Ang-2及Tie-2的表达水平与宫颈癌临床分期和有无淋巴结转移密切相关,且Ang-2、Tie-2mRNA及蛋白在宫颈癌组织中的表达明显高于正常宫颈组织,Ang-2及Tie-2可能在宫颈癌发生、发展过程中发挥重要作用。 |

| Objective To investigate the expression of angiopoietin-2 (Ang-2) and its receptor Tie-2 (Tie-2) in cervical cancer and their clinical significance. Methods The protein and mRNA expressions of Ang-2 and Tie-2 in 60 cervical cancer tissue samples and 60 normal cervical tissue samples were detected by immunohistochemical technique, RT-PCR and Western blot, respectively. The association between Ang-2, Tie-2 expression and clinical features was analyzed by Spearman analysis.
Results Ang-2 and Tie-2 were detected in the cytoplasm of cancer cells. The expression of Ang-2 and Tie-2 were significantly higher in cervical cancer tissues than that in normal tissues (all P<0.05). The expression of Ang-2 and Tie-2 was significantly higher in patients with lymph node metastasis and clinical stage III-IV than that in patients without lymph node metastasis and with clinical state I-II (all P<0.05). Expression of Ang-2 was positively correlated with expression of Tie-2 (r=0.847 9, P<0.01). Conclusion The expressions of Ang-2 and Tie-2 are up-regulated in cervical cancer, particularly in late stage cancer with lymph
node metastasis, which indicates that Ang-2 and Tie-2 may play an important role in the progress of cervical cancer. |
