		     | 目的探讨微囊蛋白1、细胞内钙离子在转化生长因子茁1(TGF-茁1)诱导哮喘大鼠气道平滑肌细胞(ASMCs)增殖中的作用。方法离体培养大鼠ASMCs,并分为正常组、哮喘组、TGF-茁1干预组、TGF-茁1+β-环糊精(茁-CD)干预组,CCK-8法检测各组细胞的增殖水平;Westernblot法检测各组微囊蛋白1含量;流式细胞仪检测各组ASMCs胞内钙离子荧光强度。结果哮喘组OD值较正常组明显增加(P<0.05),TGF-茁1组OD值较正常组、哮喘组明显增加(P<0.05),TGF-茁1+茁-CD组OD值较TGF-茁1组明显增加(P<0.05)。TGF-茁1组较正常组和哮喘组微囊蛋白1含量减少(P<0.05),TGF-茁1+茁-CD组较TGF-β1组微囊蛋白1含量减少(P<0.05)。TGF-茁1组较正常组和哮喘组胞内钙离子荧光强度增加(P<0.05),TGF-茁1+茁-CD组较TGF-茁1组胞内钙离子荧光强度增加(P<0.05)。相关关系分析示细胞增殖水平与微囊蛋白1含量呈负相关(r=-0.51,P<0.05),细胞增殖水平与钙离子荧光强度呈正相关(r=0.60,P<0.05),微囊蛋白1含量与钙离子荧光强度呈负相关(r=-0.87,P<0.05)。结论微囊蛋白1可以抑制TGF-茁1诱导的ASMCs增殖,这种抑制作用可能部分是通过减少胞内游离钙离子来实现的。 |

                | Objective    To  investigate  the  effect  of  caveolin-1  and  intracellular  Ca2+    ([Ca2+] (i))  on  TGF-β1-induced
proliferation of airway smooth muscle cells(ASMCs) in asthmatic rats. Methods ASMCs of rats were isolated and cultured in vitro. CCK-8 method was used to detect cell proliferation and Western blot method was used to detect the expression of caveolin-1 protein. FCM was used to detect [Ca2+](i). Results OD value in asthma group was higher than that in normal group (P<0.05), OD value in TGF-β1 group was higher than that in normal group and asthma group (P<0.05), OD value in TGFβ1+β-CD group was higher than that in TGF-β1 group (P<0.05). The expression of caveolin-1 in TGF-β1 group was significantly lower than that in normal group and asthma group (P<0.05), the expression of caveolin-1 in TGF-β1+ β-CD group was significantly lower than that in TGF-β1 group (P<0.05). The [Ca2+](i) of ASMCs in TGF-β1 group was higher than that in normal group and asthma group(P<0.05), the[Ca2+](i) of ASMCs in TGF-β1+ β-CD group was higher than that in TGF-β1 group(P<0.05). There was negative correlation between cell proliferation and the expression of caveolin-1 protein (r=-0.51, P<0.05), cell proliferation was positively correlated with the [Ca2+](i) (r=0.60, P<0.05), caveolin-1 protein was negatively correlated with the [Ca2+](i)(r=-0. 87,  P <0.05).     Conclusion     Caveolin-1  cab  suppress  TGF-β1-induced  ASMCs  proliferation  in  asthmatic  rats,  which  is
associated with the decrease of [Ca2+](i). |
