目的检测MMP12mRNA在人肺腺癌组织中的表达,探讨其与临床病理特征的关系。方法收集肺腺癌患者肿瘤组织样本39例,每例患者另取癌旁组织作为对照,应用real-timePCR技术,分别以GAPDH、RPLP0、PUM1、HPRT1和TBP为内参基因分析肺腺癌肿瘤组织及癌旁组织MMP12mRNA表达水平。扩大样本至103例,以PUM1为内参基因分析肺腺癌肿瘤组织及癌旁组织MMP12mRNA表达水平及MMP12mRNA表达水平与患者临床病理特征的关系。结果肺腺癌肿瘤组织相对于癌旁组织MMP12mRNA表达水平上调者所占比例较表达水平无改变或表达水平下调者增高,且以PUM1为内参基因检测肺腺癌肿瘤组织MMP12mRNA表达水平高于癌旁组织(P<0.05),且MMP12mRNA在男性患者(男性vs女性,2.07±3.25vs0.53±2.90,P<0.05)、病理亚型为实性/微乳头状生长为主浸润性腺癌(实性/微乳头状生长为主vs附壁样/乳头状/腺泡状生长为主,3.30±2.95vs0.75±3.20,P<0.05)中表达水平上调。结论MMP12mRNA在肺腺癌肿瘤组织中表达水平上调,可能参与肺腺癌发生、发展,且性别可能作为肺腺癌肿瘤组织MMP12mRNA表达水平上调的危险因素。 |

Objective To investigate the expression of MMP12 mRNA in human lung adenocarcinoma and its clinicopathological significance. Methods The expression of MMP12 mRNA was detected with real-time quantitative PCR using GAPDH, RPLP0, PUM1, HPRT1, TBP as reference genes in 39 specimens of lung adenocarcinoma and corresponding pericancerous tissues. The expression of MMP12 mRNA was also detected in 103 samples of lung adenocarcinoma tissue (including above 39 cases) with PUM1 as reference gene. The association between expression of MMP12 mRNA and clinical and pathological characteristics of lung adenocarcinoma was analyzed. Results More adenocarcinoma samples had MMP12 mRNA up-regulated than samples with not-changed or down-regulated MMP12 mRNA, and the expression of MMP12 mRNA was
significantly higher in tumor tissues than that in pericancerous tissues (P <0.05). The rate of MMP12 mRNA expression was
significantly higher in male patients than that in females(2.07±3.25 vs 0.53±2.90, P<0.05), and in invasive solid or micropapillary predominant tumors was higher than that in lepidic, papillary or acinar predominant tumors (3.30±2.95 vs 0.75±3.20, P<0.05). Conclusion The expression of MMP12 mRNA is up-regulated in lung adenocarcinoma, particularly in male patients, indicating that MMP12 may be associated with occurrence and development of lung cancer. |
