		     | 目的探讨茶多酚对人前列腺癌PC-3M细胞中调控细胞凋亡的生存素(Survivin)mRNA和蛋白表达的影响。方法在人前列腺癌PC-3M细胞培养器皿中分别加入用完全培养液稀释的0、40、60、80滋g/ml茶多酚溶液,24、48h后应用RT-PCR和Westernblot检测并比较SurvivinmRNA、蛋白表达水平。结果与0滋g/ml组相比,40滋g/ml组加入茶多酚48h后SurvivinmRNA、蛋白表达水平均明显下调(均P<0.05);60、80滋g/ml组加入茶多酚24、48h后SurvivinmRNA、蛋白表达水平均明显下调(均P<0.05),其中80滋g/ml组下调最为明显(均P<0.05);大致呈剂量、时间依赖性(均P<0.05)。结论茶多酚可下调SurvivinmRNA和蛋白表达水平,促进人前列腺癌PC-3M细胞调亡,在一定范围内呈剂量、时间依赖性。 |

                | Objective To investigate the effect of tea polyphenols on the mRNA and protein expression of survivin in prostate cancer PC-3M cells. Methods Human prostate cancer PC-3M cells were treated with 40, 60, 80滋g/ml tea polyphenol for 24h or 48h, the mRNA and protein expression of survivin in PC-3M cells were detected by RT-PCR and western blot, respectively. Results The mRNA and protein expression of survivin in PC-3M cells treated with 60滋g/ml and 80滋g/ml for 24h and 48h was significantly decreased in a dose-dependent and time-dependent manner(P<0.05). Conclusion Tea polyphenols can promote apoptosis of human prostate cancer PC-3M cells, which is related to the down-regulation of survivin gene tran- scription and translation. |
