		     | 目的研究核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)炎性体相关分子表达与老年2型糖尿病(T2DM)的关系。方法采用ELISA法测定30例老年T2DM患者、30例健康体检者(对照组)血浆中IL-1β、IL-18和IL-37水平,分别采用定量聚合酶链反应(PCR)技术和ELISA技术检测糖尿病患者单核细胞来源的巨噬细胞(MDM)表达NLPR3、凋亡相关斑点样蛋白(ASC)和IL-1βmRNA水平及葡萄糖诱导MDM分泌IL-1β和TNF-琢的水平。结果老年T2DM患者血浆中的IL-1β和IL-18水平均显著高于对照组,差异均有统计学意义(均P<0.05);而IL-37无统计学差异;T2DM患者MDM中NLPR3、ASC和IL-1βmRNA表达水平均显著高于对照组,差异均有统计学意义(均P<0.05);且经葡萄糖刺激后IL-1β、TNF-琢水平显著高于对照组,差异有统计学意义(均P<0.05)。结论NLRP3炎性体相关分子的表达与炎性体活化可能参与老年T2DM的炎症反应过程。 |

                | Objective To investigate the expression of NLRP3 inflammasome-related molecules in elderly patients with type 2 diabetes mellitus (T2DM) . Methods The plasma levels of IL-1β, IL-18 and IL-37 in 30 T2DM patients and 30 healthy controls were measured with ELISA; the expression of NLRP3, ASC and IL-1β in monocytes derived macrophages (MDM) were detected by real time PCR. The contents of IL-1β and TNF-琢 secreted by MDM treated by glucose were also analyzed. Results Plasma IL-1β and IL-18 levels in T2DM patients was significantly higher than those in healthy controls (P<0.05), while IL-37 level was no significantly increased (P >0.05). Expression of NLRP3, ASC and IL-1 in MDM and IL-1β, TNF-琢 secreted by MDM were significantly higher than those in controls (P<0.05). Conclusion The NLRP3 inflammasome pathway may be involved
in the inflammatory process of elderly patients with T2DM. |
