IFN-γ 诱导蛋白 10 及其受体在子痫前期孕妇血清和胎盘组织中的表达

摘要:
目的 探讨子痫前期孕妇血清及胎盘组织中 IFN-γ 诱导蛋白 10(IP-10)及其受体 CXCR3 的表达情况。 方法 选 择同期住院的晚期正常妊娠、轻度子痫前期和重度子痫前期孕妇各 20 例。采用 ELISA、实时荧光定量 RT-PCR 和 Western blot 等方 法检测血清 IP-10 水平、CXCR3 mRNA 表达水平,以及胎盘组织中 IP-10、CXCR3 蛋白和 mRNA 表达水平;并对子痫前期孕妇 IP-10 与 CXCR3 进行 Pearson 相关分析。 结果 与正常妊娠组比较,轻、重度子痫前期孕妇血清 IP-10 水平和 CXCR3 mRNA 表达 水平均有不同程度的增高(均 P<0.05),且均为重度子痫前期组高于轻度子痫前期组(均 P<0.05)。此外,轻、重度子痫前期孕妇胎盘 组织中 IP-10、CXCR3 蛋白和 mRNA 水平均明显高于正常妊娠组(均 P<0.05)。Pearson 相关分析显示,子痫前期孕妇血清 IP-10 水 平与外周血 CXCR3 mRNA 表达水平呈正相关(r=0.914,P<0.05);子痫前期孕妇胎盘组织中 IP-10 mRNA 表达水平与 CXCR3 mRNA 表达水平呈正相关(r=0.907,P<0.05)。 结论 IP-10 及其受体 CXCR3 表达上调参与子痫前期病程,子痫前期孕妇血清、胎 盘组织中 IP-10 与 CXCR3 均呈正相关,可能与子痫前期的发病有关。

Expression and significance of IFN-款 inducible protein 10 and its receptor CXCR3 in serum and placenta in pregnant women with preeclampsia

Abstract:
Objective To investigate the expression of IFN-γ inducible protein 10 (IP-10) and its receptor CXCR3 in placenta and serum of gravidas with preeclampsia. Methods Samples of placenta and serum were collected from normal gravidas, gravidas with mild preeclampsia and gravidas with severe preeclampsia with 20 cases in each group. The protein and mRNA expression of IP-10 and CXCR3 in placenta and serum was detected by ELISA, real-time PCR and Western blot, respectively. Results ELISA showed that serum IP-10 levels in patients with severe or mild preeclampisa were significantly higher than those in normal pregnancy group (P<0.05). The mRNA expression of CXCR3 in peripheral blood of patients with severe or mild preeclampisa was significantly higher than that in normal pregnancy group (P<0.05). Real-time PCR and Western blot showed that the expression levels of IP -10 and CXCR3 mRNA and protein in the placenta of severe or mild preeclampisa groups were significantly higher than those in normal pregnancy group (P<0.05). Conclusion The elevated expressions of IP-10 and its receptor CXCR3 in the placenta and serum may be involved in the pathogenesis of preeclampsia.
