鳕鱼皮寡肽对人胃癌细胞BGC-823增殖影响的实验研究

摘要:
目的探讨鳕鱼皮寡肽(codskinofoligopeptide,CSO)对人胃癌细胞(BGC-823)增殖的影响。方法用不同浓度(20、40、60、80mg/ml)CSO处理体外培养的BGC-823,72h后显微镜下观察细胞生长情况;分别于24、48、72h,应用CCK-8法检测细胞增殖情况并计算50%细胞抑制率(IC50);处理后24h采用流式细胞术测定细胞凋亡及细胞周期的改变情况。结果与对照组相比,经CSO处理后,镜下BGC-823数目减少。CCK-8检测结果显示,BGC-823数目减少呈剂量、时间依懒性(均P<0.05),并且CSO对BGC-823作用24、48和72h的IC50分别是87.34、68.14、44.55mg/ml。对照组BGC-823存活率为(86.62±10.32)%,60mg/ml组BGC-823存活率为(24.18±6.59)%,差异有统计学意义(P<0.05);对照组BGC-823晚期凋亡率为(1.81±0.35)%,60mg/ml组BGC-823晚期凋亡率为(55.84±5.89)%,差异有统计学意义(P<0.05)。在G1期,对照组BGC-823细胞分布为(61.13±10.32)%,60mg/ml组BGC-823细胞分布为(79.74±10.21)%,差异有统计学意义(P<0.05);在S期,对照组BGC-823细胞分布为(23.04±8.64)%,60mg/ml组BGC-823细胞分布为(10.42±5.36)%,差异有统计学意义(P<0.05);在G2期,20mg/ml组BGC-823细胞分布为(9.96±2.68)%,40mg/ml组BGC-823细胞分布为(6.93±1.25)%,60mg/ml组BGC-823细胞分布为(9.84±3.79)%,与对照组BGC-823细胞分布(15.83±5.89)%相比,差异均有统计学意义(均P<0.05)。结论CSO可阻滞BGC-823生长周期,促进BGC-823凋亡,抑制增殖,从而抑制肿瘤细胞的生长。

Effect of cod skin oligopeptide on proliferation of human gastric carcinoma BGC-823 cells

Abstract:
Objective To explore the effect of cod skin oligopeptide (CSO) on the proliferation of human gastric carcinoma BGC-823 cells. Methods BGC-823 cells were treated with different concentrations (20, 40, 60, 80mg/mL) of CSO. Cell morphology was observed under the microscope, cell viability was determined by CCK-8 assay, and ell apoptosis and cell cycle were detected by flow cytometry. Results CSO significantly suppressed the proliferation of BGC-823 cells in a time- and dose-dependent manner. IC50 of CSO with 24h, 48h and 72h were 87.34mg/ml, 68.14mg/ml and 44.55mg/ml, respectively. The early apoptosis rate of BGC-823 cells was (17.55±5.48)% and the late apoptosis rate was (55.84±5.89)%, when the cells were treated with 80mg/ml CSO. After BGC-823 cells were treated with CSO for 24h, the cell cycle arrested at S/G2 stage. The number of G1 cells increased with the increasing of CSO concentration, peaked at (79.74 ±10.21)% when CSO concentration was 60mg/ml, the number of S/G2 cells were decreased with the increasing of CSO concentration, reached the lowest value of (9.84± 3.79 )% when CSO was 60mg/ml. Conclusion CSO can inhibit the proliferation and promote the apoptosis of human gastric cancer BGC-823 cells.
