目的探讨线粒体辅酶Q(MitoQ)对高脂饮食大鼠心肌功能的作用。方法将36只8周龄雄性SD大鼠随机分为高脂+MitoQ组(高脂饲料喂养16周后,加入MitoQ继续喂养4周)、高脂组(高脂饲料喂养20周)和正常对照组(基础饲料喂养20周)各12只。先行经胸常规超声心动图检测;再应用速度向量成像(VVI)技术测量乳头肌水平左心室短轴观各个节段最大收缩期运动速度(Vs)、最大径向应变(着r)、最大切向应变(着c)、最大收缩期径向应变率(SRr)、切向应变率(SRc)等指标;实验结束后处死大鼠进行组织学检查。结果3组大鼠舒张末期室间隔厚度(IVSTd)、舒张末期左心室后壁厚度(LVPWTd)、左心室舒张末期内径(LVEDd)、左心室收缩末期内径(LVEDs)、射血分数(EF)、短轴缩短率(FS)比较,差异均无统计学意义(P>0.05)。高脂组Vs、着r、着c、SRr、SRc均较正常对照组明显下降(均P<0.05),亦低于高脂+MitoQ组(均P<0.05)。高脂组较正常对照组心肌细胞排列紊乱,肌纤维断裂,心肌间质胶原纤维明显增生;高脂+MitoQ组较高脂组心肌细胞排列规整,未见明显的肌纤维断裂,心肌间质胶原纤维减少。结论高脂饮食会损害大鼠左心室心肌收缩功能,MitoQ可起到一定的保护作用。 |

Objective To investigate the effect of MitoQ on myocardial function in high-fat diet rats. Methods A total of 36 male 8-week-old SD rats were randomly assigned to high-fat diet with MitoQ treatment group, high-fat diet group and normal diet group (n=12, in each group). Rats in control group were fed with basic diet, rats in high-fat diet group were fed with high-fat diet for 20 wks, and those in high-fat diet with MitoQ group fed with high-fat diet for 20 wks and MitoQ was added from wk16 till wk 20. Transthoracic routine echocardiography and velocity vector imaging (VVI) were performed in 3 groups, and parameters Vs, 着r, 着c, SRr and SRc were measured at papillary muscle level in left ventricular short axis view. By the end of experiments animals were sacrificed for histological examination. Results There were no significant differences in IVSTd,
LVPWTd, LVEDd, LVEDs, EF and FS among 3 groups(P >0.05). The Vs, 着r, 着c, SRr and SRc in high-fat diet group were significantly lower than those in normal control group (P<0.05) and those in high-fat diet with MitoQ treatment group (P<0.05). Compared to control group, histological examination showed that there were disorder of myocardial cells arrangement and muscle fiber fracture, significant increasing of collagen fibers in intercellular substance, and the deposition of collagen fibers in heart. However, these histopathological changes were attenuated in high-fat diet with MitoQ group. Conclusion High-fat diet can damage the myocardial systolic function of left ventricle in adult male rats, MitoQ treatment may protect the myocardial function from the high fat diet-induced damage. |
