结缔组织生长因子在肾积水模型大鼠肾组织中的表达研究

摘要:
目的观测结缔组织生长因子(CTGF)在肾积水模型大鼠肾组织中的表达,并与先天性肾积水患儿进行对比,为先天性肾积水的靶点防治研究提供可靠的动物模型。方法将48只成年健康雄性Wistar大鼠分为模型组和对照组各24只。模型组行左侧肾盂输尿管处游离并结扎输尿管,对照组仅行左侧肾盂输尿管处游离。两组在术后1、2、4、8周各随机选取6只,行肾脏病理学检查,用免疫组织化学染色及Westernblot法检测CTGF在组织中的分布及蛋白的表达水平。结果CTGF蛋白含量随着梗阻时间延长而增加。CTGF在模型组肾脏中各时点的表达明显高于对照组,差异均有统计学意义(均P<0.01)。结论CTGF在大鼠肾积水模型中为阳性表达,且与梗阻时间呈正相关,与前期先天性肾积水患儿的研究结果相一致,此模型可用于先天性肾积水的CTGF靶点防治。

Abstract:
Objective To investigate the expression of connective tissue growth factor (CTGF) in kidney tissue of rat hydronephrosis model. Methods Forty eight healthy male Wistar rats were equally divided into two groups: in model group the left pelviureteric junction was dissociated and the ureter was ligated, and in control group the left pelviureteric junction was dissociated only. Six rats in each group were sacrificed 1, 2, 4 and 8 weeks after the operation and the left kidney was taken for histopathological examination. The expression of CTGF protein and mRNA in kidney tissue was detected with immunohistochemistry and Western blot, respectively. Results The expression of CTGF mRNA and protein in model group was significantly higher than that in the control group (P<0.01) and increased with the time length of pelviureteric junction obstruction. Conclusion The expression of CTGF is upregulated in the rat hydronephrosis model and positively correlated with the obstruction duration, indicating that CTGF might be used as a target of preventing congenital hydronephrosis.
