系统性红斑狼疮患者B细胞中microRNA-155的表达及干预后效应

摘要:
目的分析microRNA-155(miR-155)在系统性红斑狼疮(SLE)患者外周血B细胞中的表达及干预后B细胞表达IgG水平。方法收集66例SLE患者及10例健康人群外周血,分离出B细胞,采用实时荧光定量PCR(qRT-PCR)检测miR-155在B细胞中的表达水平,利用电转染技术使miRNA抑制剂干预SLE患者外周血B细胞miR-155的表达,然后以ELISA检测B细胞干预后培养液上清液IgG的水平。结果qRT-PCR结果显示,与健康人比较,SLE患者外周血B细胞中miR-155表达上调(P<0.05),通过miRNA抑制剂下调SLE患者B细胞中的miR-155的表达后,B细胞分泌的IgG水平明显降低。结论SLE患者的B细胞中表达上调的miR-155可能参与了SLE患者B细胞功能调控。

Expression of miRNA-155 in peripheral B cells of patients with systemic lupus erythematosus

Abstract:
Objective To investigate the expression of microRNA-155 (miR-155) in peripheral B cells of the patients with systemic lupus erythematosus (SLE). Methods Sixty-six SLE patients and 10 healthy controls were enrolled in the study. The B cells were isolated from peripheral blood, and the expression of miR-155 in B cells was detected by fluorescence quantitative real-time RT-PCR (qRT-PCR). The levels of IgG in supernatants of B cells were determined by enzyme-linked immunosorbent assay (ELISA) after miR-155 expression was intervened by electrotransfection. Results The expression of miR-155 was significantly increased in B cells of the patients with SLE compared with healthy controls (P<0.05). After intervention of the miR-155 expression with miRNA inhibitor, the levels of total IgG in the supernatants of the cultured B cells were significantly de- creased. Conclusion The study indicates that the up-regulated miR-155 expression may be involved in the regulation of B cell function in systemic lupus erythematosus.
