乌药提取物对酒精性肝损伤大鼠门静脉内毒素及小肠组织形态学的干预研究

摘要:
目的观察乌药不同提取物对酒精性肝损伤模型大鼠门静脉内毒素(ET)及小肠组织形态学的影响。方法选取雄性SD大鼠88只随机分为8组,每组11只,分别为正常对照组、模型对照组、阳性对照组、乌药水提取物组、乌药醇提取物组、乌药醇提取物石油醚萃取物组、乌药醇提取物乙酸乙酯萃取物组、乌药醇提取物正丁醇萃取物组。除正常对照组(以等体积蒸馏水灌胃)外,其余各组大鼠均给予乙醇体积分数50%的白酒以1.2ml/100g连续灌胃33d建立酒精性肝损伤模型,造模同时给予阳性药物及乌药提取物1.0ml/100g连续灌胃,正常对照组及模型对照组给予等量蒸馏水。末次给药后禁食12h取血,测定门静脉ET、TNF-α含量;迅速处死并在冰台上摘取小肠,采用HE染色法观察小肠病理学改变,电镜观察肠黏膜微绒毛。结果(1)与正常对照组相比,模型对照组大鼠门静脉ET、TNF-α含量明显升高(P<0.01);光镜下小肠黏膜部分肠上皮细胞变性、坏死,间质轻度水肿;电镜下小肠上皮可见不规则、稀疏的微绒毛及肿胀、空泡变性的线粒体。(2)与模型对照组相比,各给药组门静脉ET、TNF-α含量均明显降低(P<0.05或0.01);光镜下小肠组织病理损伤程度明显减轻;电镜下小肠上皮呈紧密连接的簇状微绒毛增多,线粒体形态较之正常。(3)药效对比,乌药醇提取物调节门静脉血ET和炎症因子的作用优于水提取物及阳性药物;醇提取物中,醇提石油醚萃取物和醇提正丁醇萃取物效果较优。结论乌药不同提取物抗大鼠酒精性肝损伤作用机制与抑制内毒素———TNF-α通路有关,可能是通过改善酒精引起的肠黏膜损伤、减轻肠源性内毒素血症来实现。

Extracts from Lindera aggregata (Sims) Kosterm for intervention of alcoholic liver disease in rats

Abstract:
Objective To investigate the efficacy of extracts from Lindera aggregata (Sims) Kosterm in intervention of alcoholic liver disease in rats. Methods Eighty eight male SD rats were divided into 8 groups with 11 in each group: normal control group, model group, positive control group, WYSTW, WYCTW, WYCTC1, WYCTC2 and WYCTC3. The alcoholic liver disease was induced by intragastrical administration of 1.2 mL liquor (50% v/v)/100 g body weight for 33 days, at the same time the different extracts of Lindera aggregata (Sims) Kosterm were given to rats in various groups; the equal volume of distilled water was given to rats in normal control group and model group. At the end of experiments the serum endotoxin and TNF-α were examined, and intestinal tissues were assessed with HE staining. Results The serum endotoxin and TNF-α in the model group was significantly higher than those in control group (P<0.05), while it decreased in intervention groups as compared with the model group (P<0.05 or 0.01). The pathological degree of intestinal damage in model group was significantly severer than that in control group, but the intervention group was significantly lower than that of model group. Irregular, sparse microvilli and swollen and vacuolated mitochondria was observed in small intestinal epithelium in the model group, but the histology of intestine was normal in the intervention groups. The effect of alcohol extract, especially in alcohol extraction of petroleum ether extract and alcohol extraction of butyl alcohol extract of Lindera aggregata(Sims) Kosterm was superior to that of water extract in reducing the endotoxemia and serum inflammatory factors. Conclusion The alcohol induced intestinal mucosal injury and endotoxemia can be protected by the extracts of Lindera aggregata (Sims) Kosterm in rats.
