AQP1和AQP3在宫颈癌新辅助化疗前后的表达变化及意义

摘要:
目的观察新辅助化疗前后宫颈癌组织中水通道蛋白1(AQP1)与水通道蛋白3(AQP3)表达的变化,探讨两者在宫颈癌新辅助化疗中的意义。方法选择宫颈鳞癌患者30例,术前接受以顺铂为基础的有效新辅助化疗。化疗前和手术中分别取癌组织标本,应用RT-PCR法、Westernblot法和免疫组化法对水通道蛋白(AQPs)的mRNA、蛋白和组织分布进行检测。结果化疗后AQP1和AQP3的mRNA表达量约为化疗前的3倍和2.7倍,差异均有统计学意义(均P<0.05)。AQP1和AQP3蛋白在宫颈癌组织中大量表达,在细胞膜和胞质中均有分布,化疗后表达量提高,以胞质更为明显。30例患者经过化疗后,其中19例完全缓解,11例部分缓解。完全缓解组患者化疗后AQP1和AQP3的mRNA和蛋白表达的增值虽然均大于部分缓解组,但差异均无统计学意义(均P>0.05)。结论AQP1和AQP3在新辅助化疗后宫颈癌组织中的表达增加,增幅可能与化疗效果相关。

Expression of AQP1 and AQP3 in cervical cancer patients with neoadjuvant chemotherapy

Abstract:
Objective To investigate the expression of aquaporin-1 (AQP1) and aquaporin-3 (AQP3) in cervical cancer patients with neoadjuvant chemotherapy. Methods Thirty patients with cervical cancer were enrolled in the study. Effective neoadjuvant chemotherapy with cis-platinum was given in all cases before surgery. Tumor specimens were obtained before chemotherapy and during surgery. The expression of AQP1 and AQP3 mRNA and protein was determined by RT-PCR, Western blot and immunohistochemistry, respectively. Results The mRNA expression of AQP1 and AQP3 increased3 and 2.7 times after chemotherapy, respectively. The AQP1 and AQP3 proteins were expressed abundantly in both cytomembrane and cytoplasm of cancer tissue; and the protein expression increased significantly after chemotherapy, especially in cytoplasm. Among the thirty patients, 19 cases achieved complete remission (CR) and 11 cases partial remission (PR) after chemotherapy. There was no significant difference in AQP1 and AQP3 expression between CR cases and PR cases, although there was a trend of increasing expression in CR patients. Conclusion Expression of AQP1 and AQP3 in cervical cancer is increased after neoadjuvant chemotherapy.
