肝癌衍化生长因子对大肠癌早期血管形成的调节作用研究

摘要:
目的探讨肝癌衍化生长因子(HDGF)在大肠癌发生、发展过程中对血管形成的调节作用及其在大肠癌早期诊断中的意义。方法选取50例大肠癌组织及对应的癌旁正常肠黏膜组织(距癌周>10cm)标本,25例腺瘤性息肉组织标本作为实验材料。采用免疫组化法检测大肠癌、腺瘤、正常肠黏膜组织HDGF蛋白、血管内皮生长因子(VEGF)的表达及微血管密度(MVD),并进行比较。荧光定量RT-PCR检测大肠癌、腺瘤、正常肠黏膜组织HDGFmRNA的表达并进行比较。结果大肠癌、腺瘤、正常肠黏膜组织HGDF蛋白、VEGF蛋白表达阳性率及MVD比较差异均有统计学意义(均P<0.05),大肠癌组织最高,腺瘤组织次之,正常肠黏膜组织最低。大肠癌、腺瘤、正常肠黏膜组织HDGFmRNA表达水平比较差异均有统计学意义(均P<0.05),大肠癌组织>腺瘤>正常肠黏膜组织(均P<0.05)。结论HDGF在大肠癌组织中高表达,通过与VEGF协同作用,促进大肠癌早期血管形成。HDGF表达在大肠癌恶变过程中呈递增趋势,可将其作为大肠癌早期诊断的辅助指标。

Expression of hepatoma-derived growth factor (HDGF) in colon carcinoma

Abstract:
Objective To investigate the expression of hepatoma-derived growth factor (HDGF) in colon carcinoma and its significance. Methods Fifty tissue samples of colon carcinoma and the adjacent normal mucosa, and 25 samples of adenomatous polyp were collected. The expression HDGF, vascular endothelial growth factor (VEGF) and microvessel density (MVD) in tissue samples were detected by immunohistochemical method; the expression of HDGF mRNA was detected by real-time RT-PCR. Results There were significant differences in positive rate of HGDF and VEGF protein expression and MVD among colon carcinoma, adenoma, and normal intestinal mucosa (P<0.05). These indicators were highest in carcinoma, followed by those in adenoma, and those in normal intestinal mucosa were the lowest. The expression of HDGF mRNA in colon carcinoma was the highest, followed by that in adenoma, and that in normal intestinal mucosa was the lowest (P<0.05). Conclusion The expression of HDGF is up-regulated along with the expression of VEGF and MVD in colon carcinoma; and also the expres- sion of HDGF shows an increasing trend from normal mucosa, adenomatous polyps to carcinoma of the colon.
