沉默β-catenin表达对胃癌MGC-803细胞生物学行为的影响与作用机制研究

摘要:
目的探讨β-链蛋白(β-catenin)表达下调对胃癌MGC-803细胞生物学行为的影响,并分析其作用机制。方法将RNA干扰慢病毒(β-catenin-RNAi慢病毒)及空载体慢病毒(β-catenin-Neg)分别感染MGC-803细胞,分为实验组(MGC-803-RNAi细胞)、阴性对照组(MGC-803-Neg细胞)、空白对照组(未处理的MGC-803细胞)。Westernblot检测验证转染效率;双染流式细胞术检测细胞凋亡率,单染流失细胞术分析细胞周期,Transwell法检测细胞侵袭能力;Westernblot检测细胞Bax、Bcl-2、cyclinD1及基质金属蛋白酶(MMP)-7蛋白表达水平。结果MGC-803-RNAi、MGC-803-Neg细胞中显示绿色荧光的细胞约占80%,MGC-803细胞不显示荧光;与MGC-803-Neg、MGC-803细胞相比,MGC-803-RNAi细胞β-catenin表达水平明显下调(P<0.05),细胞凋亡率升高(P<0.05),出现G1/S期阻滞现象(P<0.05),侵袭能力明显减弱(P<0.05),Bcl-2、cyclinD1及MMP-7蛋白表达水平下调(均P<0.05),而Bax蛋白表达水平上调(P<0.05)。结论沉默β-catenin表达可促进胃癌MGC-803细胞凋亡,阻滞其细胞周期进程,减弱其侵袭转移力,其作用机制可能与影响Wnt/β-catenin信号通路下游靶蛋白表达有关。

Abstract:
Objective To investigate the effect of silencing β-catenin gene expression on biological behaviors of gastric carcinoma cells and the related molecular mechanisms. Methods Human gastric cancer MGC-803 cells were infected with β-catenin-RNAi lentivirus (study group) and β-catenin-negative lentivirus (negative control group), respectively. The expression of β-catenin, Bax, Bcl-2, cyclin and MMP-7 proteins was detected by Western blotting. The apoptosis, cell cycle and migration of infected and non-infected (normal control group) MGC-803 cells were determined by flow cytometry and Transwell assay, respectively. Results The expression levels of β-catenin in study group was lower than that of the other two groups(P<0.05). Cell apoptosis was increased(P<0.05), cell cycle was blocked at G1/S phase(P<0.05) and the migration ability was inhibited in MGC803 cells after infected with β-catenin-RNAi lentivirus (P<0.05). The expression of Bcl-2, cyclin D1 and MMP-7 proteins was inhibited but the expression of Bax was increased in MGC cells after β-catenin expression was silenced. Conclusion The silence of β-catenin gene expression can lead to increasing cell apoptosis, inhibiting cell cycle and migration ability in gastric cancer MGC-803 cells, which are associated with the down-regulation of f Bax, Bcl-2, cyclin D1 expression and up-regulation of MMP-7 expression.
