目的探讨辅助性T细胞(Th)17在过敏性哮喘发病中的作用及其与Notch信号通路的关系。方法选择过敏性哮喘患儿40例(哮喘组)和体检儿童40例(健康对照组)。采用流式细胞仪检测其Th17细胞比例,实时荧光定量聚合酶链反应检测外周血单个核细胞Notch1、Notch4、维甲酸相关孤儿核受体(RORγt)mRNA表达水平,ELISA法检测外周血清IL-17水平。过敏性哮喘患儿Notch1mRNA表达水平分别与Th17细胞比例、RORγtmRNA表达水平和IL-17水平进行相关性分析。结果哮喘组Th17细胞比例、Notch1及RORγtmRNA表达水平、IL-17水平均显著高于健康对照组(均P<0.05),而两组儿童Notch4mRNA表达水平比较差异无统计学意义(P>0.05)。哮喘组Notch1mRNA表达水平与Th17细胞比例、RORγtmRNA表达水平、IL-17水平均呈正相关(r=0.780、0.555和0.636,均P<0.01)。结论Th17细胞在过敏性哮喘中发挥重要作用,Notch1可能通过影响Th17表达参与过敏性哮喘的病理过程。 |

Objective To investigate the role of T helper cell 17 (Th17) in the pathogenesis of allergic asthma and its relation to Notch signaling pathway. Methods Forty children with allergic asthma and 40 healthy children (controls) were enrolled in the study. Flow cytometry was used to detect the percentage of Th17cell. Quantity real-time polymerase chain reaction (qRT-PCR)was used to
detect the mRNA expression of Th17 related transcription factor retinoid-related orphan nuclear receptor γt (RORγt), the mRNA
expression of Notch1 and Notch4 in peripheral blood mononuclear cells (PBMC). The concentrations of IL-17 in plasma were tested by enzyme linked immunosorbent assay (ELISA). The correlations of Notch1 mRNA expression with the percentage of Th17 cell,
RORγt expression in PBMC and plasma IL-17 concentration were analyzed. Results There was a significantly increased
percentage of Th17 cells in allergic asthmatic children compared to healthy controls(P<0.01). The mRNA expression of RORγt was significantly higher in allergic asthmatic children compared to healthy controls(P<0.01). The plasma level of IL-17 was significantly higher in asthmatic children (P<0.05). There was a significant increase of the mRNA expression of Notch1, but no increase in the mRNA expression of Notch4 (P >0.05). The mRNA expression of Notch1 was positively correlated with the percentage of Th17 cells, RORγt expression and plasma IL-17 levels(r=0.780, 0.555 and 0.636, all P<0.01). Conclusion Th17 may play an important role in the pathogenesis of allergic asthma in children, which may be associated with the Notch1 signaling pathway. |
