		     | 目的探讨三结构域蛋白(TRIM)1、occludin、ZO-1在结肠癌组织中的表达及临床意义。方法采用免疫组化法检测120例结肠癌患者手术切除的癌组织及其癌旁正常组织中TRIM1、occludin、ZO-1表达,分析TRIM1与occludin、ZO-1表达的相关性,以及TRIM1、occludin、ZO-1表达与结肠癌患者临床特征及预后的关系。结果与癌旁正常组织比较,结肠癌组织TRIM1表达升高,occludin、ZO-1表达降低,差异均有统计学意义(均P<0.05)。结肠癌组织中TRIM1表达与occludin、ZO-1表达均呈负相关(rs=-0.420、-0.343,均P<0.05)。结肠癌组织中TRIM1、occludin、ZO-1表达与肿瘤临床分期、分化程度、T分期、是否淋巴结转移等有关(均P<0.05),与患者性别、年龄、肿瘤部位等均无关(均P>0.05)。经生存分析,TRIM1高表达者5年累积生存率明显低于TRIM1低表达者(P<0.05),occludin高表达者5年累积生存率明显高于occludin低表达者(P<0.05),ZO-1高表达者5年累积生存率明显高于ZO-1低表达者(P<0.05)。TRIM1高表达是影响结肠癌患者术后5年总生存的独立危险因素(HR=2.374,95%CI:1.04~5.44,P<0.05)。结论结肠癌组织中TRIM1表达升高,且与occludin、ZO-1表达呈负相关;3项指标均与结肠癌的临床分期、分化程度、T分期、是否淋巴结转移等有关,其中TRIM1高表达是影响患者术后5年总生存的独立危险因素。 |

                | Objective To investigate the expression and clinical significance of TRIM1, occludin and ZO-1 in colon cancer. Methods The expression of TRIM1, occludin and ZO-1 protein in cancer and paracancerous tissues of 120 patients undergoing colon cancer resection was detected by immunohistochemistry. The correlation of TRIM1 with occludin and ZO-1 protein expression, and the relationship between TRIM1, occludin, ZO-1 protein expression and clinicopathological parameters and prognosis of patients were analyzed. Results Compared with normal adjacent tissues, TRIM1 expression in colon cancer increased, occludin and ZO-1 expression decreased(P<0.05). TRIM1 expression was negatively correlated with occludin and ZO-1 expression in colon cancer tissues (r=-0.420, -0.343, P<0.05). In colon cancer tissues, the expression of TRIM1, occludin, ZO-1 expression was correlated with clinical stage, differentiation degree, T stage, lymph node metastasis(P< 0. 05 ), not correlated with gender, age and tumor site (P>0.05). The 5-year cumulative survival rate was significantly lower in patients with high expression of TRIM1 than that in those with low expression of TRIM1(P<0.05), while the 5-year cumulative survival rate in patients with high ZO -1 and occlu din expression was significantly higher than that in those with low ZO -1 and occludin expression (P<0.05). High TRIM1 expression was an independent risk factor for 5 years postoperative overall survival in patients
with colon cancer (HR=2.374, 95%CI:1.04-5.44, P<0.05). Conclusion In colon cancer, the expression of TRIM1 is up-regulated, while the expression of occludin and ZO-1 is down-regulated. High TRIM1 expression would independently affect 5-year overall survival of colon cancer patients after operation. |
