Treg/Th17对小鼠非酒精性脂肪性肝炎的影响研究

摘要:
目的探讨Treg/Th17对小鼠非酒精性脂肪性肝炎(NASH)的影响,以期为寻找NASH的新治疗靶点提供参考。方法将20只4~6周龄SPF级雄性C57BL/6小鼠按随机数字表法分为模型组和对照组,各10只。对照组小鼠予普通饮食饲养10周。模型组小鼠予高脂饮食饲养10周,建立小鼠NASH模型。10周后处死两组小鼠留取外周血与肝组织。观察并比较模型组与对照组小鼠肝组织病理学变化,血清TG、ALT、AST水平,外周血Treg/Th17比例及肝组织炎症因子水平。结果模型组小鼠肝细胞具备NASH的HE染色和Masson染色特征,非酒精性脂肪性肝病活动度评分、肝纤维化评分均高于与对照组(均P<0.05)。模型组小鼠血清TG、ALT、AST水平均高于对照组(均P<0.05)。与对照组比较,模型组小鼠促炎因子NF-资B、IL-6、IL-17水平均升高(均P<0.05),而抗炎因子IL-10水平下降(P<0.05)。结论Treg/Th17失衡会引起促炎因子与抗炎因子之间的失衡,促进小鼠NASH与肝纤维化的形成。

Treg/Th17 ratio in mouse non-alcoholic steatohepatitis

Abstract:
Objective To investigate the Treg/Th17 ratio in nonalcoholic steatohepatitis(NASH) in mice. Methods Twenty C57BL/6 mice were randomly divided into control group and model group. The NASH was induced by feeding high-fat diet for 10 weeks in model group, and the control group was given conventional diet. Peripheral blood and liver tissue samples were acquired after mice were sacrificed after 10 weeks. Liver histopathological changes, serum levels of TG, ALT and AST, the ratio of Treg/Th17 in peripheral blood and the level of inflammatory factors in liver tissue were compared between the two groups. Results The model group had the characteristics of NASH in tissue sections with HE and Masson staining. The NASH activity score (NAS), liver fibrosis score(FibroS) and biochemical indexes including TG, ALT, AST in model group were significantly higher than those in control group (P<0.05). And the expression of proinflammatory cytokines NF-kB, IL-6 and IL-17 were increased significantly (P<0.05), while the expression of anti-inflammatory factor IL-10 decreased significantly (P<0.05). Conclusion The inflammation and fibrosis in mouse NASH are associated with the imbalance between pro inflammatory factors and anti-inflammatory factors, which may be resulted from the change of Treg/Th17 ratio.
