5-氨基酮戊酸光动力抗感染化学疗法对中型无绿藻抑制作用的体外研究

摘要:
目的观察体外5-氨基酮戊酸光动力抗感染化学疗法对中型无绿藻的抑制作用。方法实验组分为3组:1、5、10mmol/L5-氨基酮戊酸溶液分别与中型无绿藻37℃避光孵育30min后接受激光照射20min,即T1、T2、T3组;对照组分为3组:C1组为不加5-氨基酮戊酸溶液的中型无绿藻37℃避光孵育30min后不接受照射,C2组为加5-氨基酮戊酸溶液的中型无绿藻37℃避光孵育30min后不接受照射,C3组为不加5-氨基酮戊酸溶液的中型无绿藻37℃避光孵育30min后接受照射。采用菌落计数法分析5-氨基酮戊酸光动力抗感染化学疗法对中型无绿藻的抑制作用。结果7株中型无绿藻3个对照组菌落计数比较,差异均有统计学意义(均P<0.05);经两两比较,C2组菌落计数均明显低于C1组、C3组(均P<0.05)。3个实验组每株中型无绿藻生长均受到抑制,与C2组菌落计数比较差异均有统计学意义(均P<0.05)。仅有3株中型无绿藻(O22a、O22c、O22d)3个实验组菌落计数比较,差异均有统计学意义(均P<0.05);另4株中型无绿藻(O22b、O22e、O22f、O22g)3个实验组菌落计数比较,差异均无统计学意义(均P>0.05)。以C2组为参照,计算不同浓度5-氨基酮戊酸光动力处理后中型无绿藻菌生长总的受抑制率,T1、T2和T3组分别为(59.09±10.27)%、(70.44±11.01)%和(70.90±7.12)%,合并后总的受抑制率为41.78%~87.82%,平均为(66.81±10.94)%。T1、T2和T3组中型无绿藻菌生长总的受抑制率比较,差异有统计学意义(P<0.05);经两两比较,T2、T3组中型无绿藻生长受抑制率均明显高于T1组(P<0.05),T2与T3组比较差异无统计学意义(P>0.05)。结论体外5-氨基酮戊酸溶液光动力抗感染化学疗法对中型无绿藻具有明显抑制作用。

Photodynamic effect of 5-aminolevulinic acid on growth of prototheca zopfii in vitro

Abstract:
Objective To evaluate the effect of 5-aminolevulinic acid (ALA) on growth of prototheca zopfii in vitro. Methods The suspensions of prototheca zopfii were incubated with ALA at concentrations of 1mmol/L (T1), 5mmol/L (T2), or 10mmol/L( T3 ) at 37℃ in dark for 30min, then were irradiated with narrow band LED( 633 ± 10 ) nm at the light intensity of 100mW/cm2 and at a distance of 1cm for 20min. The following controls were included, spore suspensions in saline without irradiation or ALA(C1), spore suspensions in saline with 5mmol/L ALA without irradiation(C2), and spore suspensions in saline with irradiation without ALA (C3). Aliquots of suspensions from each group were inoculated on Sabouraud dextrose agar, which was incubated at 37℃ for 48-96h, the numbers of developing colonies were counted. Results There was significant difference in growth of prototheca zopfii between the control groups C1, C2 and group C3 (P<0.05). The growth of prototheca zopfii was significantly reduced in ALA-photodynamic treatment groups (T1, T2 and T3) compared to the control group(C2) (P<0.05). There was significant difference in growth among the ALA-photodynamic treatment groups in three isolates(O22a, O22c, O22d), while no significant difference in 4 isolates (O22b, O22e, O22f, O22g). Growth reduction was (59.09±10.27)%, (70.44±11.01)% and (70.90±7.12)% in T1, T2 and T3 treatment groups respectively. There was significant difference of growth reduction between T1 and the other two groups (P<0.05), no significant difference between T2 and T3 group (P>0.05). In vitro study shows ALA-PDT induced 41.78%-87.82% reduction of the growth of all tested prototheca zopfii strains. Conclusion Our study demon strates that ALA photodynamic therapy can suppress the growth of prototheca zopfii in vitro.
