FTY720对蛛网膜下腔出血大鼠海马神经元凋亡的抑制作用

摘要:
目的探讨FTY720对蛛网膜下腔出血大鼠海马神经元凋亡的抑制作用。方法将48只大鼠随机分成正常组、假手术组、模型组和治疗组,每组12只;采用枕大池二次注血法制作蛛网膜下腔出血大鼠模型。治疗组大鼠按1mg/kg腹腔注射FTY720,另外3组大鼠腹腔注射0.9%氯化钠注射液1ml。24h后处死大鼠,采用原位末端标记(Tunel)法检测海马组织凋亡神经细胞,Westernblot法检测海马组织原癌基因(c-fos)、pro-Caspase-3和pro-Caspase-9蛋白表达,四肽荧光底物法检测海马组织Caspase-3、Caspase-9蛋白活性;比较4组大鼠以上指标的差异。结果正常组与假手术组大鼠凋亡的海马神经元比例,c-fos、pro-Caspase-3、pro-Caspase-9蛋白表达水平以及Caspase-3、Caspase-9蛋白活性比较,差异均无统计学意义(均P>0.05);模型组较假手术组、正常组均明显升高(均P<0.05);治疗组较模型组均明显下降(均P<0.05)。结论FTY720可明显抑制蛛网膜下腔出血大鼠海马神经元凋亡,具有脑保护作用。

Inhibitory effect of FTY720 on apoptosis of hippocampal neuron in rats with subarachnoid hemorrhage

Abstract:
Objective To investigate the inhibitory effect of FTY720 on apoptosis of hippocampus neuron in subarachnoid hemorrhage rats. Methods Forty eight rats were randomly assigned to normal group, sham-operation group, model group and treatment group with 12 rats in each group. The rat model of subarachnoid hemorrhage was established by double autologous blood injection into cisterna magna. Rats in treatment group were administrated intraperitoneally with 1mg/kg FTY720. Rats in other three groups were given an intraperitoneal injection of 1ml normal saline. After 24 hours, rats were sacrificed. Apoptotic nerve cells in hippocampal tissue were detected by TUNEL, the expression of c-fos, pro-caspase-3 and pro-caspase-9 proteins was detected by Western blot, and the activity of caspase-3 caspase-9 proteins in hippocampal tissues was detected by tetrapeptide fluorescence substrates. The differences of the above indexes were compared among the four groups. Results There were not significant differences in the number of apoptotic neuronal cells, expressions of c-fos, pro-Caspase-3 and pro-Caspase-9 proteins as well as activities of Caspase-3 and Caspase-9 proteins between normal group and sham-operation group (all P >0.05). Compared with sham-operation group and normal group, the above variables were significantly elevated in rats of model group (all P <0.05). In comparison with model group, the above variables were markedly decreased in rats of treatment group. Conclusion FTY720 exerts a suppressive effect on apoptosis of hippocampus neuron in subarachnoid hemorrhage rats and possesses a cerebral protective effect.
