		     | 目的研究维生素D受体(VDR)基因启动子甲基化异常与婴儿尿路感染的关系。方法选择急性尿路感染患儿32例(尿路感染组)和同期健康体检婴儿30例(健康对照组)。采用荧光定量PCR法检测VDR、DNA甲基化转移酶(DNMTs)mRNA表达水平,重亚硫酸氢钠测序技术检测VDR基因启动子的甲基化水平。结果尿路感染组患儿VDRmRNA表达水平高于健康对照组婴儿,DNMT1mRNA表达水平低于健康对照组婴儿,两组比较差异均有统计学意义(均P<0.05)。两组DNMT3a、DNMT3bmRNA表达水平比较差异均无统计学意义(均P>0.05)。尿路感染组患儿VDR基因启动子甲基化水平显著低于健康对照组婴儿,两组比较差异有统计学意义(P<0.05)。结论尿路感染患儿VDRmRNA表达水平升高,VDR基因启动子的甲基化水平降低,VDR基因甲基化异常与婴儿尿路感染相关。 |

                | Relationship between abnormal methylation of vitamin D receptor gene promoter and urinary tract infection in infants |

                | Objective To analyze the relationship between methylation of vitamin D receptor(VDR) gene promoters and urinary tract infection (UTI) in infants. Methods Thirty two infants aged 1~12 months with UTI (UTI group) and 30 normal infants with matched  ages ( control group) were enrolled  in  the  study. The mRNA  expressions  of VDR  and  DNA  methyltransferase
(DNMT1, DNMT3a, DNMT3b) were detected by real-time PCR; the methylation levels of VDR gene promoters were detected by bisulfite sequencing(BSP). Results The expression of VDR mRNA in UTI group was higher than that in control group(P<0.05). The value of DNMT1 mRNA in UTI group was lower than that in control group(P<0.05). There was no significant difference in expression levels of DNMT3a mRNA and DNMT3b mRNA between two groups (both P >0.05). The methylation of VDR gene promoter in UTI group was significantly lower than that in control group(P<0.05).    Conclusion    The expression  level  of  VDR
mRNA is up-regulated in patients with UTI, while the methylation of VDR gene promoter is decreased, abnormal methylation of VDR gene is associated with urinary tract infection in  infants. |
