目的 探讨溴结构域蛋白4(bromodomain-containing protein 4,BRD4)作为胃癌诊断标志物的可能,并探究其与免疫浸润的相关性。 方法 生物信息学分析BRD4在胃癌组织和正常胃黏膜组织中的表达水平,与胃癌患者预后之间的关系。受试者工作特征(receiver operating characteristic curve,ROC)曲线用于评估BRD4对胃癌的诊断价值,并进一步探讨BRD4表达水平与胃癌患者临床病理因素之间的相关性。基因探针富集分析(gene set enrichment analysis,GSEA)揭示BRD4激活的信号通路。通过RT-qPCR和Western blot分别检测正常胃黏膜上皮细胞和胃癌细胞中BRD4 mRNA和蛋白的表达水平。 结果 生物信息学分析和细胞实验均证实BRD4的mRNA和蛋白水平均在胃癌中高表达(P<0.001),其高表达与患者预后不良显著相关。BRD4区分胃癌组织和正常组织的曲线下面积高达0.850。GSEA表明BRD4与调节免疫反应的通路密切相关。 结论 BRD4有望成为胃癌诊断的潜在生物标志物,其高表达可能诱导胃癌免疫逃逸。 |

The diagnosis value of BRD4 in stomach adenocarcinoma and its correlation with immune infiltration |

Objective To explore the possibility of bromodomain-containing protein 4 (BRD4) as a diagnostic marker for stomach adenocarcinoma (STAD) and to investigate the correlation between BRD4 and immune infiltration. Methods Bioinformatics analyze the levels of BRD4 in normal gastric mucosa tissues and STAD tissues, as well as the relationship between BRD4 mRNA levels and the prognosis of patients with STAD. The receiver operating characteristic (ROC) was used to assess the diagnostic value of BRD4 for STAD. Further, the correlation between BRD4 levels and clinicopathological factors of patients with STAD was explored. Gene set enrichment analysis (GSEA) revealed the signaling pathway activated by BRD4. RT-qPCR and Western blot were used to detect the mRNA and protein expression levels of BRD4 in normal gastric mucosal epithelial cells and gastric cancer cells, respectively. Results Both bioinformatics analysis and cell experiments confirmed that BRD4 was highly expressed in STAD (P<0.001). The high levels of BRD4 in patients with STAD was significantly associated with poor prognosis. The area under the curve of BRD4 in distinguishing between STAD tissues and normal tissues was as high as 0.850. GSEA indicated that BRD4 was closely related to the pathways regulating immune responses. Conclusion BRD4 is expected to be a potential biomarker for diagnosing STAD, and its high expression may induce immune escape of STAD. |
