let-7b-5p和miR-15b-5p用于评估颈动脉斑块稳定性的临床意义

摘要:
目的 探究可用于评估颈动脉斑块稳定性的循环miRNAs。方法 选择颈动脉斑块患者6例和5例同期健康体检者作为健康对照组,采用miRNA芯片分析颈动脉粥样硬化患者的循环miRNAs表达谱;选择低回声颈动脉斑块患者30例和强回声颈动脉斑块患者30例,采用TaqMan实时定量PCR方法检测miRNAs 的表达水平并作比较;采用ROC曲线评估miRNAs用于区分不同类型颈动脉斑块的诊断效能。结果 采用miRNA芯片共发现41个差异表达的循环miRNAs,其中23个表达上调,18个表达下降。低回声颈动脉斑块组中let-7b-5p和miR-15b-5p的表达水平均明显高于强回声颈动脉斑块组(P值分别为0.013和0.002)。let-7b-5p和miR-15b-5p用于区分低回声和强回声颈动脉斑块的AUC值分别为(0.730,0.787)。结论 let-7b-5p和miR-15b-5p有望作为评估颈动脉斑块稳定性的生物标志物。

Clinical value of let-7b-5p and miR-15b-5p for assessment of carotid atherosclerotic plaque

Abstract:
Objective: To explore circulating miRNAs for assessment of carotid atherosclerosis plaque stability. Methods: 6 patients with carotid atherosclerotic plaque and 5 healthy subjects were collected for miRNA microarray analysis to screen differentially expressed miRNAs. 30 patients with hypoecho carotid plaque and 30 patients with hyperecho carotid plaque were collected for comparation of the expression levels of miRNAs by TaqMan quantitative PCR. ROC curve analysis was performed to determine the diagnostic utility. Results: Via miRNA microarray analysis, a total of 41 differentially expressed miRNAs were profiled, including 23 up-regulated and 18 down-regulated miRNAs. The expression levels of let-7b-5p and miR-15b-5p in hypoecho carotid plaque group were all significantly higher than those in hyperecho carotid plaque group (P =0.013, 0.002, respectively). For discrimination of hypoecho and hyperecho carotid plaque, the area under the curve of let-7b-5p and miR-15b-5p was 0.730 and 0.787, respectively. Conclusion: This study suggested that let-7b-5p and miR-15b-5p may serve as potential biomarkers for assessment of plaque stability.
