| 目的 探讨S-亚硝基谷胱甘肽(SNG)对脓毒症大鼠急性肺损伤(ALI)的保护作用。方法 采用盲肠结扎和穿孔术(CLP)建立脓毒症模型,将大鼠随机分为假手术组(Sham)、Sham+SNG组、CLP 组和 CLP+SNG组,每组10只。采用苏木精-伊红(H&E)染色和肺湿/干重比评估肺损伤程度,检测支气管肺泡灌洗液(BALF)中总蛋白和细胞水平,以及肺组织中肿瘤坏死因子-α(TNF-α)、白细胞介素- 1β(IL-1β)、IL-6和IL-17A的水平,同时检测肺组织中谷胱甘肽(GSH)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶的水平。结果 SNG显著提高了CLP大鼠的生存数量(P < 0.05),降低了脓毒症大鼠肺组织损伤评分(P < 0.001)、肺湿/干重比(P < 0.001)以及BALF中总细胞数、总蛋白、中性粒细胞数和淋巴细胞数(均P < 0.001),并降低肺组织中TNF-α、IL-1β、IL-6和IL-17A的水平(均P < 0.001)。同时,SNG显著提高脓毒症大鼠肺组织中抗氧化酶GSH、SOD、GSH-Px 和过氧化氢酶的水平(均 P < 0.001)。结论 SNG对脓毒症大鼠急性肺损伤具有显著保护作用,具体机制与抗炎和抗氧化相关。 |

| Objective To investigate the protective effect of S-nitrosoglutathione (SNG) on acute lung injury (ALI) in septic rats. Methods? A rat model of sepsis was established by cecal ligation and puncture (CLP). The rats were randomly divided into four groups: sham operation group (Sham), Sham + SNG group, CLP group, and CLP + SNG group, with 10 rats in each group. Hematoxylin-eosin (H&E) staining and lung wet/dry weight ratio were used to evaluate the degree of lung injury. The levels of total proteins and cells in bronchoalveolar lavage fluid (BALF) were detected. Additionally, the levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), IL-6, and IL-17A in lung tissues were measured. Meanwhile, the levels of glutathione (GSH), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase in lung tissues were detected. Results ?SNG significantly increased the survival number of CLP rats (P < 0.05), and reduced the lung tissue injury score (P < 0.001), lung wet/dry weight ratio (P < 0.001), as well as the total cell count, total protein, neutrophil count, and lymphocyte count in BALF (all P < 0.001) in septic rats. It also decreased the levels of TNF-α, IL-1β, IL-6, and IL-17A in lung tissues (all P < 0.001). Moreover, SNG significantly increased the levels of antioxidant enzymes including GSH, SOD, GSH-Px, and catalase in the lung tissues of septic rats (all P < 0.001). Conclusion ?SNG exerts a significant protective effect on acute lung injury in septic rats, and its specific mechanism is related to anti-inflammation and antioxidation. |
