目的探讨过氧化物酶体增殖物激活受体-γ2(PPAR-γ2)基因Pro12Ala及C161T位点单核苷酸多态性与金华市汉族人群冠心病易感性及血脂水平的关系。方法采用聚合酶连反应-限制性片段长度多态性方法对263例冠心病患者和207例健康体检者进行PPAR-γ2基因Pro12Ala及C161T位点单核苷酸多态性检测,并探讨基因多态性对冠心病发生及血脂水平的影响。结果Pro12Ala位点多态性检测显示,两组间基因型分布及等位基因频率差异均无统计学意义(均P>0.05),在冠心病组中Ala等位基因携带者TC和LDL-C水平均高于非携带者(均P<0.05)。C161T位点多态性检测结果显示,基因型和等位基因频率在两组间分布差异均无统计学意义(均P>0.05);两组T等位基因携带者TG水平低于非携带者(P<0.05)。结论PPAR-γ2基因Pro12Ala及C161T位点基因多态性可能与金华地区汉族人群冠心病发生无关,但Pro12Ala位点的Ala等位基因可能影响冠心病患者TC和LDL-C水平,C161T位点的T等位基因可能影响人群TG水平。 |

Objective To explore the association between peroxisome proliferator activated receptor-gamma 2 (PPAR-γ2) variants and risk of coronary heart disease (CHD) in Jinhua Han populations. Methods A total of 263 patients with CHD and 207 healthy controls were recruited. The Pro12Ala and C161T polymorphisms of PPAR-γ2 gene were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The association of Pro12Ala and C161T polymorphisms of PPAR-γ2 gene with CHD and plasma lipid levels was analyzed. Results Neither the frequencies of genotypes nor frequencies of alleles of Pro12Ala polymorphisms of PPAR-γ2 gene was statistically different between CHD group and control group (P >0.05), but subjects with PA/AA genotypes had a higher plasma mean concentration of TC and LDL-C compared with those with the PP genotype in the CHD group (P<0.05). There were also no significant differences in the distribution of C161T genotypes and alleles between CHD and control groups(P >0.05), but subjects with CT/TT genotypes had a lower plasma mean TG concentration than those with CC genotype(P<0.05). Conclusion There may be no association between
PPAR-γ2 gene Pro12Ala and C161T polymorphisms with CHD in Jinhua Han population, but the CHD patients who had PA/AA genotypes of Pro12Ala polymorphisms may have higher plasma levels of TC and LDL-C, and people who had CT/TT genotypes of C161T polymorphisms may have lower plasma level of TG. |
