SMURF1、RUNX3 在甲状腺乳头状癌组织中的表达及其临床意义

摘要:
目的 探讨 SMURF1、RUNX3 蛋白在甲状腺乳头状癌(PTC)中的表达及其与患者临床病理特征的相关性。 方法 采 用免疫组化 SP 法检测 60 例 PTC 组织及 20 例正常甲状腺组织中 SMURF1、RUNX3 蛋白的表达,分析两者在 PTC 与正常甲状腺组 织中的表达差异及其与 PTC 患者临床病理特征的相关性。 结果 PTC 组织中 SMURF1 蛋白阳性表达率高于正常甲状腺组织,而 RUNX3 阳性表达率低于正常甲状腺组织(均 P<0.01);SMURF1、RUNX3 蛋白的表达与 PTC 患者的 TNM 分期、病理分级及淋巴结转 移均相关(均 P<0.05),而与患者性别、年龄及肿瘤大小等因素均无关(均 P >0.05)。PTC 组织中 SMURF1 的蛋白表达与 RUNX3 蛋 白表达呈负相关(r=-0.564,P<0.05)。 结论 SMURF1、RUNX3 蛋白在 PTC 组织中表达异常,并呈负相关,提示两者可能相互作 用,共同促进 PTC 发生、发展,两者可作为 PTC 分类和分期的辅助指标。

Abstract:
Objective To investigate the expression of SMURF1 and RUNX3 in papillary thyroid carcinoma and its clinicopathological significance. Methods Immunohistochemical SP method was applied to detect the expression of SMURF1 and RUNX3 in 60 samples of papillary thyroid carcinoma (PTC) and 20 samples of normal thyroid tissue. The relationship of SMURF1, RUNX3 expression with clinical and pathological features of PTC was analyzed. Results The positive rate of SMURF1 protein expression in PTC were significantly higher than that in normal thyroid tissue, while the positive rate of RUNX3 protein expression in PTC were significantly lower(P<0.01). The expression of SMURF1 and RUNX3 in PTC was correlated with TNM stage, histological grade and lymph node metastasis of PTC (P<0.05), however, was not correlated with sex, age of patients and tumor size(P >0.05). SMURF1, RUNX3 protein expression in PTC was negatively correlated with each other(r=-0.564, P<0.05). Conclusion The abnormal expression of SMURF1 and RUNX3 in papillary thyroid carcinoma suggests that these two proteins might be used as auxiliary clinicopathological indicators of the cancer.
