血液灌流对急性百草枯中毒兔肝组织炎症损伤的影响

摘要:
目的观察血液灌流对急性百草枯中毒兔肝组织炎症损伤影响,并探讨可能的机制。方法将60只普通级雄性日本大耳白兔按随机数字表法分为4组,正常对照组6只,空白对照组18只,百草枯染毒组18只,血液灌流(HP)治疗组18只。正常对照组给予0.9%氯化钠溶液50mg/kg灌胃,空白对照组给予0.9%氯化钠溶液50mg/kg灌胃1h后给予HP治疗持续2h,百草枯染毒组给予20%百草枯50mg/kg灌胃染毒,HP治疗组百草枯灌胃染毒1h后给予HP治疗持续2h。于第1、3、7d麻醉处死,并留取肝脏。检测血浆ALT、AST和肝组织TBIL、TNF-α、IL-6和NF-κB基因和蛋白的表达。光镜下观察肝脏组织病理改变。结果与正常对照组相比,空白对照组各观察指标无变化,百草枯染毒组血浆AST、ALT、TBIL和肝组织TNF-α、IL-6、NF-κBmRNA和蛋白表达量在第1、3、7天明显升高(P<0.05)。与百草枯染毒组相比,HP治疗组血浆AST、ALT、TBIL和肝组织TNF-α、IL-6、NF-κBmRNA和蛋白表达量在第1、3、7天显著下降(P<0.05)。光镜下观察,百草枯染毒组兔肝组织病理损害在染毒后第3天最严重,在第1、3、7天,HP治疗组肝脏病理损害较百草枯染毒组轻。结论HP治疗可能够减轻百草枯对家兔肝脏的炎症损伤,对百草枯中毒所致的兔肝脏损伤具有保护作用。

Hemoperfusion alleviates paraquat-induced liver inflammatory injury in rabbit

Abstract:
Objective To investigate the effect of hemoperfusion (HP) on liver inflammatory injury induced by paraquat in rabbits. Methods Sixty male rabbits were randomly divided into 4 groups:normal control group(n=6) received gavage with 0.9% saline 50mg/kg,blank control group (n=18) received gavage with 0.9% saline 50mg/kg followed by HP for 2h, paraquat group (n=18) received gavage with 20% paraquat 50mg/kg, HP group (n=18) received gavage with 20% paraquat 50mg/kg followed by HP for 2h. On d 1, 3, 7 the rabbits were sacrificed in batches, and the liver samples were collected. The serum levels of AST, ALT and TBIL were measured, the expressions of TNF-α, IL-6, NF-κBmRNA and NF-κB protein and mRNA in liver tissues were detected by BSA and RT-PCR, respectively. Results Compared with the normal control group,the levels of AST, ALT, TBIL, TNF-α, IL-6, NF-κBmRNA , NF-κB protein were increased in paraquat group (P<0.05) . Compared with paraquat group, the AST, ALT, TBIL,TNF-α, IL-6,NF-κBmRNA ,NF-κB protein levels in HP group were significantly decreased (P <0.05).The pathological results of liver tissues showed that no abnormalities were found in normal control and blank control groups, the morphological injury was severe in paraquat group, which was alleviated in HP group. Conclusion HP significantly increase resistance to Paraquat-induced inflammation injury in the rabbit liver and exert a protective effect on Paraquat-induced liver injury.
