目的探索理想的小鼠人胰腺癌原位移植瘤模型建立方法和移植瘤生长检测方法。方法将小鼠人胰腺癌皮下移植瘤制成瘤体,原位包埋于健康的BALB/c雌性小鼠胰腺尾部被膜下,20只小鼠均接种成功,建模4周后应用MicroPET检测小鼠体内移植瘤的生长情况。结果MicroPET检测显示所有小鼠人胰腺癌原位移植瘤模型均建立成功,成功率为100%;检测到肿瘤对18FDG的平均标准摄取值为(4.30±0.35)。结论瘤体原位包埋法是建立小鼠人胰腺癌原位移植瘤模型较理想的方法,操作简便,成功率高;MicroPET是检测移植瘤生长情况的可靠辅助检查方法,可以用于胰腺癌动物模型的监测。 |

Objective To establish an orthotropic transplatiation mofel of human pancreatic carcinoma in nude mouse.
Methods The small piece of human pancreatic carcinoma from xenograft was implanted subcapsularly in the tail of pancreas in
20 female BALB/c nude mice. The growth of transplanted tumors were observed with microPET imaging 4 weeks after operation, then the animals were sacrificed by cervical dislocation and the samples of tumor were obtained. Results Four weeks after implantation, the orthotopic transplatation nude mice model was established with a success rate of 100.0%. MicroPET showed that the mean uptake value of 18FDG in tumor mass was (4.30±0.35). Conclusion The orthotopic human pancreatic cancer nude mouse model has been successfully established, and MicroPET can be used to examine the growth of transplanted pancreatic carcinoma in nude mice. |
