目的探讨IL-6/sIL-6R复合物对人脐静脉内皮细胞(HUVECs)血管内皮生成因子(VEGF)及基质金属蛋白酶-9(MMP-9)表达的影响。方法采用不同浓度的IL-6(0、10、20、40、80ng/ml)联合sIL-6R(100ng/ml)刺激HUVECs,培养24h后,分别采用real-timePCR及Westernblot检测MMP-9、VEGF的mRNA及蛋白表达。结果在基因水平IL-6/sIL-6R复合物可诱导内皮细胞VEGF及MMP-9mRNA的表达;在蛋白水平IL-6/sIL-6R复合物亦可促进VEGF及MMP-9的表达。结论IL-6/sIL-6R复合物上调血管内皮细胞VEGF及MMP-9的表达,在类风湿关节炎(RA)的发病机制中起着重要的作用。 |

Abstract: |
Objective To investigate the regulatory effect of IL-6/sIL-6R complex on expression of MMP-9 and VEGF in human umbilical vein endothelial cells (HUVECs). Methods Cultured HUVECs were treated with sIL-6R (100ng/ml) and various concentrations of IL-6 (0, 10, 20, 40 and 80ng/ml) for 24h, the expression of MMP-9 and VEGF was detected by real-time fluorescent quantitative PCR and Western blot. Results The mRNA and protein expressions of MMP-9 and VEGF were increased in presence of IL-6/sIL-6R complex (P <0.05). Conclusion IL-6/sIL-6R complex can promote the expression of MMP-9 and VEGF in human umbilical vein endothelial cells, indicating that IL-6/sIL-6R complex may be involved in the pathogenesis of rheumatoid arthritis. |
